摘 要:目的通过灌肠治疗的方法观察臭氧对糖尿病大鼠模型视网膜中超氧化物岐化酶(SOD)、丙二醛(MDA)及一氧化碳(NO)的作用从而探讨臭氧对糖尿病大鼠视网膜氧化应激的作用。方法取鼠龄为12~16周清洁级SD雄性大鼠40只,其中10只做为空白对照,余30只使用STZ诱导形成糖尿病大鼠模型,标准为2次空腹血糖≥11.1 mmol/l视为造模成功(24只成功);取10只为模型组,继续高脂高糖饮食1周后改为普食;而余14只同模型组饮食,但给浓度为50μg/kg的臭氧混合气体灌肠治疗,每周3次,共计1个月;随后在腹腔注射麻醉下取眼球,剥离视网膜,-80℃冷冻后用制作成均浆,分别按SOD、MDA及NO试剂盒说明进行相应的检测。结果 30只造模大鼠中6只死亡;模型组与臭氧治疗组大鼠在体重及血糖的差异上无统计学意义(P〉0.05);SOD活力在模型组与臭氧组较空白组降低(F=6.804,P〈0.05),以模型组低于空白组为著(P〈0.05),臭氧组SOD活力较模型组稍有提高但无统计学差异(P〉0.05);相对于空白组MDA含量在模型组与臭氧组均有不同程度的升高(F=4.457,P=0.05),但臭氧治疗后MDA含量较模型组稍有下降,且模型组同空白组及臭氧组同模型组间的差异均有统计学意义(P〈0.05);NO含量空白组高于其他组,但无统计学差异(F=3.463,P〉0.05),而臭氧组NO含量低于空白组有统计学意义(P〈0.05)。结论臭氧灌肠对糖尿病大鼠体重及血糖无明显的影响;臭氧可以不同程度的增加SOD活力,降低MDA的含量,从而起到减轻视网膜氧化应激的作用。但对于NO的影响尚需要进一步研究。Objective To investigate the effects of ozone on retinal levels of SOD,MDA and NO in diabetes rats and to explore an potential treatment for early stage DR. Methods Forty SD rats were used in this study. Animals in control group( n = 10) were fed with regular diet. Other rats( n = 30) were used to create diabetic models induced with STZ. Fasting blood glucose more than 11. 1mmol / l at two or more measurements was an indication of successful modeling.Some of the diabetic rats( n = 10) were continually given high sugar and high fat diet for 1 week and then changed to normal diet. Fourteen diabetic rats received ozone enema at 50 μg / kg three times per week for 4 weeks. Animals were then sacrificed and enucleated to collect retina tissue. Results Diabetes were established in totally 24 rats. There was no difference in blood sugar or body weight between diabetic rats that treated with ozone and that not treated with ozone. Retinal SOD activity decreased in all diabetic rats when compared with normal rats( F = 6. 804,P〈0. 05). Decreased SOD activity was more profound in untreated diabetic rats than in ozone treated rats but the difference was not significant. MDA increased in all diabetic rats( F = 4. 457,P〈0. 05),and was even higher in rats treated with ozone than in untreated rats( P〈0. 05). NO content was lower in diabetic rats but the change was not significant. However,NO was significantly lower in rats treated with ozone than in normal rats( P〈0. 05). Conclusion Ozone has no effect on blood glucose and body weight in diabetic rats. Ozone can increase SOD activity,mildly decrease MDA in diabetic retina. But its effect on NO is still uncertain.
