游泳运动对非酒精性脂肪肝大鼠血清Irisin及PPARα蛋白表达的影响

摘　　要：目的:通过实验研究游泳运动对非酒精性脂肪肝大鼠血清Irisin及肝组织PPARα的蛋白表达水平相关性,探究运动对非酒精性脂肪肝的影响及相关的调节机制。方法:将40只雄性SD大鼠分为普通饲料组10只(C组),高脂膳食组30只(H组),12周非酒精性脂肪肝模型建立成功。将非酒精性脂肪肝模型大鼠随机分为非酒精性脂肪肝对照组(NC组)、非酒精性脂肪肝有氧运动组(NE组),每组10只。NE组大鼠进行为期6周的游泳训练,6天/周。结果:C组肝脏形态正常,颜色鲜红,NC组肝脏体积明显增大,颜色不鲜艳红润,NE组有改善;与C组相比,NE组血清Irisin蛋白表达量显著升高(P<0.05)。结论:运动促使Irisin表达上升,Irisin表达水平影响肝组织PPARα的蛋白表达,进而干预改善大鼠非酒精性脂肪肝病,可以考虑作为独立于减肥对非酒精性脂肪肝的调节途径,成为新的治疗靶点。Objective： To investigate the effects of swimming exercise on Irisin,PPAR alpha protein expression in serum of nonalcoholic fatty liver rats and to find out the impact and adjustment mechanism of exercises on non-alcoholic fatty liver. Methods： 40 male SD rats are randomly selected among which 10 are fed with normal fodder are recorded as group C and 30 are fed with only high-fat fodder are marked as group H. After 12 weeks,nonalcoholic fatty liver disease model is established. We then divided rats in non-alcoholic fatty liver model into non-alcoholic fatty liver comparison group（ Group NC） and non-alcoholic fatty liver aerobic group（ NE group） with 10 rats in each group. Rats in NE group are trained with swimming exercise for 6 weeks and 6 days per week. Results： Rats in Group C appear to have a normal liver shape with fresh red color while an obvious increase in volume can be seen among rats in group NC with dark red color. A notable improvement can be seen in NE group. Compared with group C,the expression level of Irisin protein in serum is greatly increased（ P 0. 05）. Conclusion： Exercises will help increase the expression level of Irisin which will impact the expression of PPARαproteins in liver tissues and thus help cure non-alcoholic fatty liver diseases of rats. We suggest it be considered as an adjustment measure for non-alcoholic fatty liver and another treatment target different from weight control.
