摘  要:目的观察腧穴针刺对偏头痛大鼠中脑P物质(SP)及其受体NK-1 m RNA表达的影响,为探讨针刺治疗急性偏头痛机制提供依据。方法 40只健康Wistar雄性大鼠按随机数字表法分为空白对照组(A组)、模型对照组(B组)、腧穴电针组(C组)、伪电针组(D组)4组,每组10只。各组按设计方案进行造模,和针刺预防治疗后,断头取中脑,应用实时定量PCR技术,测定各组大鼠中脑P物质及其受体NK-1 m RNA的表达量。结果与A组比较,B、C、D组SP m RNA、NK-1 m RNA表达量明显增高(P<0.05)。与B组比较,C、D组SP m RNA、NK-1 m RNA表达量明显降低(P<0.05)。与D组比较,C组SP m RNA、NK-1 m RNA表达量明显降低(P<0.05)。结论针刺能通过下调急性期偏头痛大鼠中脑SP及NK-1基因表达水平,发挥预防治疗偏头痛的作用,且腧穴针刺疗效更佳。Objective:To observe the effect of electroacupuncture on the expression of substance P(SP) and its receptor Neurokinin-1 m RNA in the Mesencephalon in rats with migraine.Methods:40 adult male Wistar rats were equally randomized into 4 groups:the blank group(group A),the model(group B),acupoints electroacupuncture(group C),and non-acupoints electroacupuncture(group D),10 in each group.After modeling and electroacupunture treatment,all rats were decapitated,and Mesencephalon was removed and SP and receptor Neurokinin-1(NK-1) m RNA expression were detected by real-time quantitative PCR.Results:Compared with A group,the expression of SP m RNA and NK-1 m RNA rose obviously in the other three groups(P 0.05).Compared with B group,the expression of SP m RNA and NK-1 m RNA dropped obviously in group C and D(P 0.05).Compared with D group,the expression of SP m RNA and NK-1 m RNA of group C dropped obviously(P 0.05).Conclusion:Acupoints electroacupuncture can efficiently prevent and treat migraine headache by downregulating gene expression level of SP and NK-1 m RNA,and acupoints electroacupuncture has a better effect.
