摘 要:目的将随机引物应用于黄热病毒核酸检测,为口岸快速准确地检测黄热病毒提供技术支持。方法采用随机引物和特异性引物进行逆转录,分别用自行设计的引物、探针和参考文献的引物、探针,用实时荧光PCR方法对6份黄热病毒阳性尿液样本进行黄热病毒核酸检测,并对检测结果进行比较。结果与特异性引物逆转录相比,随机引物逆转录的实时荧光PCR Ct值差别不显著(F值=0.11,P>0.05)。结论随机引物可直接应用于黄热病毒检测,也可在口岸传染病监测中发挥其他作用。Objective To detect yellow fever virus nucleic acid by using random primer,and to provide technical support to rapid detection of yellow fever virus. Methods A total of 6 urine samples of 3 imported cases of yellow fever infection were detected by Real-time PCR assay. The random primer and specific primer were used to reverse transcript,Real-time PCR assay was conducted by using primers and probe designed and referenced,respectively. The Ct values had been analyzed. Results The amplification efficiency of Real-time PCR reactions by using random primer in reverse transcription had no significant difference to that by using specific primer (F value=0.11,P〉 0.05). Conclusion Random primer could be used to detect yellow fever virus directly,and had some other important roles in infectious diseases surveillance at ports.
