摘 要:目的:对半仿生提取法制备"脉复生合剂"工艺进行优化并评价新工艺的合理性,为改进该医院制剂的生产提供实验依据。方法:以阿魏酸含量和总固形物为考察指标,采用正交实验优化半仿生提取工艺,对有效成分、稳定性及毒性进行检测以评价新工艺。结果:最佳提取工艺条件为第一煎煮时加入8倍药材量、p H为4的水,第二煎煮时加入8倍药材量、p H为7.0的水,第3煎煮时加入8倍药材量、p H为8.0的水,每煎提取2.5小时,共提取7.5小时;工艺改进后制剂稳定性优于常规水提取工艺,其最大耐受剂量大于15.0 g/kg体重,属无毒级。结论:采用半仿生提取法制备脉复生工艺合理,操作简便,适用于推广生产。Objective To optimize the semi-bionic extraction technology of Maifusheng mixture and evaluate the reasonability of this advanced technique, and provide experiment basis for improving the production of hospital preparations. Method Semi-bionic extraction technology was optimized by orthogonal experiment with the contents of ferulic acid and total solids as indicators and the new process was evaluated by detecting effective composition, stability and toxicity. Result The optimal extraction method was adding 8 folds of water with pH 4 at the first decoction time, 8 folds of water with pH 7 at second time and 8 folds of water with pH 8 at the third time, 2.5 hours every time and the total time was 7.5 hours. The preparation stability of improved process was better than the conventional water extraction technology, the maximum-tolerated dose was greater than 15.0 g/kg·bw and belongs to non-toxic substance. Conclusion The semi-bionic extraction technique for preparing Maifusheng is reasonable, easy operation and can be widely applied in popularization and production.
