桑皮止咳颗粒制备工艺及止咳平喘实验研究

摘　　要：目的:对桑皮止咳颗粒制备工艺进行最佳优化,观察研究不同浓度的桑皮止咳颗粒止咳平喘的药理作用。方法:采用正交试验法,分别以挥发油提取量、桔梗皂苷D为考察指标,优选桑皮止咳颗粒的最佳提取工艺;采用小鼠"浓氨水法"、豚鼠"喷雾法"分别选用磷酸可待因和氨茶碱作为阳性药物对照,比较观察其在止咳、平喘等方面的作用。结果:挥发油最佳提取工艺为加水14倍量,提取2次,每次2 h;醇提最佳工艺为10倍量70%乙醇提取60 min。对豚鼠咳嗽潜伏期显著延长、小鼠咳嗽次数显著减少。结论:桑皮止咳颗粒的提取和制备工艺简便、可靠,重复性良好。药效学实验表明,该药对小鼠"浓氨水法"诱发的咳嗽具有明显的抑制作用,对乙酰胆碱、组织胺混合液喷雾造成的豚鼠哮喘具有明显的对抗作用(P<0.05)。Objective：To optimize the preparation technology of the Sangpi Zhike Granules and observe the pharmacological effect of the different concentration of drug on relieving cough and asthma. Method：By orthogonal test method,the preparation technology of Sangpi Zhike Granules was optimized with the determination of volatile oil content and platycodin D as the indexes. Codeine phosphate and aminophylline were chosen as positive drug,respectively. The pharmacological effect on relieving cough and asthma was observed by the mice＂concentrated ammonia water method＂and guinea pig＂spray method＂. Result：The optimum extraction technology of volatile oil is as follow：adding 14 times amount of water extracting twice,each time for 2 h. The optimum ethanol extraction technology is as follow：adding 10 times amount of 70% ethanol extracting 60 min. the granule could obviously prolong the latent period of cough for the guinea pig. Meanwhile,the times of cough decreased for the mice. Conclusion：The extraction and preparation technology of Sangpi Zhike Granules is safe,reliable,and good repeatability. Pharmacological results showed that Sangpi Zhike Granules has obvious inhibitory effect on mice＂concentrated ammonia water＂induced cough meanwhile,has obvious antagonism on acetylcholine and histamine mixed liquid spray caused by asthma in guinea pig（P〈0.05）.
