摘 要:利用电沉积法在聚碳酸酯(PC)喷金模板上制备了Pt-Ni合金纳米管,将甲胎蛋白抗体固定在由Pt-Ni合金纳米管修饰的玻碳电极上,研制成无电子媒介的甲胎蛋白免疫传感器。实验考察了pH值和培育时间的影响,在优化实验条件下该免疫传感器测定甲胎蛋白,甲胎蛋白浓度在20~100ng/mL的范围内有良好的线性关系,检出限为0.38ng/mL。传感器具有电流响应高、灵敏度好、性能稳定等优点。方法用于血样中甲胎蛋白的测定,获得较好结果。An immunosensor for direct amperometric determination of α-1-fetoprotein(AFP) was constructed by α-1-fetoprotein antibody(anti-AFP) immobilized onto a glass carbon(GC) electrode modified with Pt-Ni alloy nanotubes,which is prepared in PC spray gold template by using electrical deposition method. The effect of pH and absorption time on the electrochemical behavior of biosensor was studied. Under the optimized experimental conditions,it was successfully used to determine AFP. The current was linearly dependent on AFP concentration in the range of 20-100 ng/mL,and the frequency shifts detection limit was 0.38 ng/mL. The experimental results showed that,this method had good stability, high current response and sensitivity, etc.
