摘 要:目的观察万古霉素和穿心莲内酯联用对金黄色葡萄球菌(金葡菌)生物膜形成的影响。方法体外构建金葡菌生物膜模型;XTT减低法评价万古霉素和穿心莲内酯联用对金葡菌生物膜初始黏附能力的影响;镜下观察药物对金葡菌生物膜的形态学影响。结果 1 000、500、250 mg/L的穿心莲内酯和各个浓度(2 MIC、1 MIC、1/2 MIC、1/4 MIC、1/8MIC)的万古霉素对金葡菌生物膜的早期黏附有抑制作用;1 MIC万古霉素和穿心莲内酯联合的抑制作用比单用万古霉素强;250 mg/L的穿心莲内酯和1 MIC万古霉素联用使细菌生物膜形态结构被显著破坏。结论一定浓度的穿心莲内酯和万古霉素对金葡菌生物膜的形成和黏附均有抑制作用,且二者恰当联用可能具有协同作用。Objective To observe the effects of vancomycin in combination with andrographolide on the biofilm formation of Staphylococcus aureus. Methods The in vitro model of Staphylococcus aureus biofilms was constructed. XTT reduction assay was used to evaluate the effects of vaneomycin in combination with andrographolide on initial adhesion capability of Staphylococcus aureus biofilm. Microscope was used to observe the morphology of the biofilm after treatment, Results Andrographolide inhibited the early adhesion of Staphylococcus aureus at 1 000, 500 and 250 mg/L when combined with different concentrations of vancomycin (2 MIC, 1 MIC, 1/2 MIC, 1/4 MIC, and 1/8 MIC). The concentration of 1 MIC vancomycin in combination with andrographolide inhibited adhesion significantly stronger than vancomycin alone. Andrographolide 250 mg/L plus 1 MIC vancomycin significantly destroyed the morphology and structure of the biofilms. Conclusions Andrographolide plus vancomycin at certain concentration can inhibit the biofilm formation and adhesion of Staphylococcus aureus. Their appropriate combination is likely to have a synergistic effect
