摘 要:本文通过观察脾气虚模型大鼠及经四君子丸治疗后空肠组织中P物质的含量及其mRNA表达水平和小肠推进率的变化,探讨脾气虚状态下空肠运动功能的变化与机制。实验把SPF级雄性SD大鼠24只随机平均分为3组:对照组、模型组和治疗组。造模成功后取材,用碳末推进法测定大鼠小肠推进率,ELISA测定大鼠空肠组织P物质含量的变化,RT-PCR测定大鼠空肠组织P物质的mRNA表达变化。实验测得模型组大鼠体重、进食量显著减少,小肠推进率明显降低,P物质的含量及mRNA表达水平明显降低(P<0.05);而治疗组大鼠体重、进食量显著增加,小肠推进率显著增加,P物质的含量及mRNA表达水平明显增加(P<0.05)。实验表明脾气虚模型大鼠空肠组织中P物质的含量和mRNA表达水平降低,四君子丸对其具有调节作用。To explore the effect of Sijunzi pill (SJZP) on the expression of substance P (SP) in jejunum, the level of mRNA and intestinal propulsive rate in spleen-qi deficiency rats,24 male spleen-qi deficiency SD rats were induced and randomly divided into the model group,SJZP group and the control group (8 per group). Carbon propelling was applied to observe the intestinal propulsion rate. Enzyme-linked immuno sorbent assay (ELISA) was applied to observe the SP in jejunum and the expression of mRNA level with reverse transcription-pelymerase chain reaction (RT-PCR). In the SJZP treatment group, the SP and mRNA of splecn-qi deficiency rats were both increasing significantly (P 〈 0.05 ). The small intestinal propulsion rate in SJZP group was higher than that of the control group. Compared with the control group, the body weight, diet and drinking amount in spleen-qi deficiency group increased greatly ( P 〈 0.05 ). The decrease of SP and mRNA in the mesenteric plexus of jejunum of rat model had some relationship with the spleen-qi deficiency. The regulation on SP and mRNA were the mechanisms of SJZP for spleen-qi deficiency.
