摘  要:目的:分析酸模、皱叶酸模、巴天酸模、长刺酸模、毛脉酸模、羊蹄酸模的psbA-trnH条形码序列,探讨酸模属植物的鉴定新方法。方法:对24份植物样品提取基因组DNA,PCR聚合酶链式反应扩增psbA-trnH序列进行双向测序,序列经过Condon Code Aligner拼接后,采用MEGA 6.0软件进行序列比对,计算种内种间K2P遗传距离,构建系统邻接树(NJ Tree)。结果:表明酸模属psbA-trnH序列种内平均K2P距离均小于种间平均K2P距离;应用相似性搜索法分析结果表明psbA-trnH序列可准确鉴定酸模属植物;NJ树显示除巴天酸模和皱叶酸模聚为一支,其余酸模属植物表现出良好的单系性。结论:psbA-trnH序列可用于鉴定酸模属植物,为酸模属植物的临床安全用药提供依据。Objective: To identify Rumex acetosa,Rumex crispus,Rumex patientia,Rumex gmelinii and Rumex japonicus using psbA-trnH barcodes. Methods: Genomic DNA from twenty-four samples was extracted and the psbA-trnH regions were amplified sequenced. The genetic distances were calculated using MEGA 6. 0 in accordance with Kimura 2-parameter( K2P) model and the neighbor-joining( NJ) phylogenetic tree was constructed. Results: The results revealed that the intraspecific genetic distances of Rumex L.were lower than the interspecific genetic distances. The identification efficiency of psbA-trnH barcode using BLAST1 was high. The NJtree showed that Rumex L. could be identified according to their monophyly except Rumex patientia and Rumex crispus. Conclusion:Psb A-trn H barcode can be used to identify Rumex L. The study provides scientific foundation for the clinical safe use of traditional Chinese medicines.
