摘 要:目的探讨猕猴模拟失重后脊髓中c-fos基因mRNA和蛋白表达的变化。方法选用15只雄性猕猴,随机分为3组:模拟失重组,模拟失重后恢复组以及对照组。通过HE染色法观察脊髓颈膨大及腰膨大组织学的改变,通过免疫组织化学染色及实时荧光定量PCR(Q-PCR)技术检测3组猕猴脊髓颈膨大及腰膨大中c-fos基因的表达情况。结果模拟失重组及失重后恢复组脊髓颈膨大及腰膨大神经元较对照组皱缩,组织间隙增宽,恢复组较失重组有所减轻;FOS蛋白在脊髓颈膨大及腰膨大3组中均呈阳性表达,主要表达于神经胶质细胞;失重组及恢复组中脊髓颈膨大和腰膨大c-fos mRNA表达量较对照组有所增加,但差异无明显统计学意义。结论模拟失重引起猕猴颈段及腰段脊髓一定程度的组织学损伤,终止负荷后有所恢复,对c-fos基因的表达无明显影响。Objective To investigate the effect of simulated weightlessness on mRNA and protein expression of c-fos in spinal cord of rhesus monkey. Methods Fifteen healthy young male rhesus monkeys were randomly divided into three groups: simulated group( simulated weightlessmess 6 weeks),recovery group( 5 weeks after 6week simulated weightlessness) and control group. HE staining was used to observe the histopathological changes of cervical enlargement and lumbar enlargement of spinal cord. And the protein and mRNA expression of c-fos in spinal cord were detected by immunohistochemisty( IHC) and quantitative real time PCR( QPCR). Results Compared with the control group,histopathological examination revealed neuron shrinkage and dilated tissue spaces in simulated group and recovery group,and the pathological changes in recovery group were lighter than those in simulated group. IHC staining showed that the expression of FOS protein was detected in the three groups of cervical enlargement and lumbar enlargement of spinal cord,which expressed mostly in neuroglia cells. The expression of c-fos mRNA in simulated group and recovery group of cervical enlargement and lumbar enlargement of spinal cord were higher than that in the control group,but there was no significant difference. Conclusion Medium or long term simulated weightlessness can induce the destruction of spinal cord structure,which can be recovered after reloading to a certain extent. And the effect of simulated weightlessness on the expression of c-fos in spinal cord is unconspicuous.
