摘  要:目的观察以破血化瘀、填精补髓法组方的中药汤剂对脑出血模型大鼠热休克蛋白70(HSP70)和低氧诱导因子-1α(HIF-1α)表达的影响。方法将80只Wistar雄性大鼠随机分为4组:假手术组,脑出血模型组(模型组),破血化瘀、填精补髓法中药汤剂组(方剂组),脑血康对照组(对照组),每组20只。采用尾状核注射自体血方法制作大鼠脑出血模型,评估大鼠神经功能状态,方剂组和对照组均在大鼠麻醉清醒后开始给药干预。应用免疫组织化学法检测各组大鼠HSP70和HIF-1α表达差异。结果与模型组比较,破血化瘀、填精补髓汤剂和脑血康胶囊治疗后的脑出血模型大鼠神经功能评分显著改善,同时2组脑组织中的HSP70较模型组明显增加、HIF-1α较模型组明显减少(P<0.05,P<0.01)。各项指标方剂组均优于对照组,差异具有统计学意义(P<0.01)。结论破血化瘀、填精补髓中药汤剂可通过调节HSP70和HIF-1α保护脑出血后继发损伤的脑组织。Objective To observe the effect of the decoction of activating blood and removing stasis and supplementing essence and marrow on the expression of HSP70 and HIF-1 α in intracerebral hemorrhage (ICH) rats. Methods Eighty Wistar male rats were randomly divided into four groups: sham operation group, model group, control group and prescription group, with twenty rats in each group. The rat model of ICH was made by injecting autoIogous blood into eaudate nucleus, and the neurological function of rats was evaluated. Rats in prescription group and control group were given drug intervention after waking up. The expression of HSP70 and HIF- 1 α were detected by immunohistoehemistry. Results The neurological function scores of rats in the prescription group and the control group were significantly improved, the expression of HSPT0 was also increased significantly, the expression of HIF-1 α was reduced significantly, the test results of the prescription group were better than the control group, the difference was statistically significant. Conclusion The decoction of activating blood and removing stasis and supplementing essence and marrow can regulate the HSP70 and HIF-1 α to reduce secondary injury after cerebral hemorrhage.
