摘 要:【目的】探究番茄不孕病毒(TAV)脱除对怀白菊叶绿素含量和氮代谢水平的影响,为怀白菊脱毒苗的大田推广应用提供理论依据。【方法】以前期获得的TAV脱除怀白菊试管苗为试材,以未经脱除TAV的试管苗为对照(CK),对两者进行继代培养,并测定培养过程中叶片叶绿素含量及氮代谢的相关生理指标。【结果】继代培养过程中,CK与TAV脱除怀白菊试管苗叶片的叶绿素α和叶绿素b含量随培养天数的增加而升高,两者均以继代第22 d时的增幅最大,分别为5.24%和41.69%。与CK相比,TAV脱除提高了相同培养天数时叶片的叶绿素a、叶绿素b含量,且差异达显著(P<0.05,下同)或极显著(P<0.01)水平。怀白菊脱除TAV病毒后,其硝酸还原酶(NR)活力、游离脯氨酸和可溶性蛋白含量在继代培养过程中均显著高于CK,三者最大增幅分别为27.99%(继代22 d)、28.10%(继代22 d)和5.89%(继代38 d)。【结论】TAV脱除能有效提高怀白菊的叶绿素含量及氮代谢水平,利于其生长发育。[Objective]In order to provide theory basis for the field popularization and application of tomato aspermy virus (TAV)-free Chrysanthemum morifolium Ramat. Huaibai plantlets, the effect of TAV-free on its chlorophyll content and nitrogen metabolism were investigated. [Method ]The acquired previously TAV-free test-tube plantlets of Huaibai were taken as experimental materials, non virus-free test-tube plantlets of Huaibai were taken as control(CK), chlorophyll content and some indexes related to nitrogen metabolism in leaves were determined during subculture. [Result]During subculture, the contents of chlorophyll a and chlorophyll b in the leaves of CK and TAV-free Huaibai test-tube plantlets were into'eased with the days of culture, the increase rates of which reached up to the highest values (5.24% and 41.69%) at 22 days of sub culture, respectively, and the chlorophyll a and chlorophyll b contents of TAV-free test-tube plantlets were increased at the same time of subculture compared with CK, with significant difference (P〈0.05, the same below) or extremely significant difference(P〈0.01 ). Furthermore, the nitrate reductase(NR) activity, free proline and soluble protein contents of TAV-free Huaibai were all significantly higher than those of CK, and the increase rates of which were 27.99% (subculture of 22 d), 28.10%(subcuhure of 22 d) and 5.89%(subculture of 38 d), respectively. [Conclusion]TAV-free could effectively improve chlorophyll content and nitrogen metabolism level of Huaibai, and are conducive to its growth and development.
