摘 要:【目的】研究四指马鲅消化道黏液细胞的发育规律,为指导四指马鲅育苗过程中适时更换饵料及制定投饵策略、完善育苗工艺和提高育苗成活率提供参考依据。【方法】利用组织石蜡切片和阿利新兰-高碘酸雪夫试剂(AB—PAS)染色,观察研究1~30日龄四指马鲅消化道黏液细胞的发生过程。【结果】四指马鲅的食道最早在5日龄时出现Ⅱ型黏液细胞;9日龄仔鱼食道黏液细胞数量有所增加,胃部已有贲门部、幽门部、胃体部和盲囊部的明显区域分化,黏膜皱褶丰富,各区域大部分的黏膜表面均有Ⅰ型黏液细胞分布;15日龄仔鱼胃腺区域出现Ⅱ型黏液细胞,肠道前、中、后段均出现Ⅱ型黏液细胞,且以圆形为主;17日龄仔鱼幽门盲囊区域出现Ⅱ型黏液细胞;23和30日龄仔鱼消化道黏液细胞以Ⅱ型为主,数量持续增加。四指马鲅消化道各段黏液细胞密度、大小及分泌能力均随日龄的增加而增加,且同日龄仔鱼消化道中以食道的黏液分泌能力最强。【结论】四指马鲅消化道黏液细胞的发生和分布特征与仔鱼摄食能力日益增强及消化道结构逐渐完善的趋势相适应。[Objective]The present experiment was conducted to investigate development law of mucous ceil in digestive tract of Eleutheronema tetradactylum, in order to provide reference basis for timely changing feed stuff, formulating feeding strategy, optimizing breeding process and rising survival rate of fry. [Method]The development of mucous cell in digestive tract of E. tetradactylum from newly hatched larvae to 30 days after hatching was investigated using the tissue paraffin section and AB-PAS(Alieian blue and periodic acid Schiff reagent) staining technology. [Result]The results showed that, the mucous cell first occurred in esophagus of 5-day-old larvae, which was classified as type ]], and its number was increasing at 9 day old, at the moment, the stomach differentiated to cardiac stomach, pyloric stomach, fundi stomach and gastric caecum with mucosal coat covering surface where a large number of Ⅰ-type mucous cell were found. Ⅱ-type mucous cell was found in gastric gland of 15-day-old larvae as well as anterior, mid and posterior sections of intestine, and a majority of Ⅱ-type mucous cells were round in shape. Ⅱ-type mucous cell first appeared in pylorie caecum of 17-day-old larvae. Mucous cells were found in digestive tract of 23- and 30-day-old larvae, which were mainly Ⅱ- type, and its number increased continuously. The density and size of mucous cells as well as secretion capacity of different section of digestive tract increased with the increase of day age, and the secretion capacity in esophagus was the strongest in larvae at same day old. [Conclusion]The occurrence and distribution characteristics of mucous cells in digestive tract of E. tetradactylum is closely related to improvement of feeding ability and perfection of digestive tract.
