摘 要:Shrunken2基因编码葡萄糖焦磷酸化酶的大亚基,是玉米淀粉合成途径中的限速酶,shrunken2是Shrunken2的隐性突变等位基因,对甜玉米品质和风味的形成起着重要的作用。本研究以玉米B73的Shrunken2基因为参考序列,在58份甜玉米自交系中进行shrunken2基因的重测序,共获得了7 635 bp的核苷酸序列,涵盖了20个外显子,17个内含子以及3'UTR区域。基因多态性分析发现,在供试群体中共有多态性位点105个,包括78个SNP和27个Indel。与普通玉米B73的Shrunken2序列之间存在58个多态性位点差异,包括50个SNP和8个Indel,其中位于编码区的SNP位点包含2个同义突变和7个非同义突变。根据shrunken2基因全长序列和编码区序列可将该群体分成14种和4种单倍型。中性检测表明该基因经历了自然和人工双重选择过程。Shrunken2 encodes the large subunits of ADP-glucose pyrophosphorylase (AGPase), which catalyzes the rate limiting step in the process of starch biosynthesis in maize. While shrunken2 is the recessive mutant of Shrunken2, it plays an important role in formation of flavor and quality in sweet corn. In this research, we re-sequenced shrunken2 gene in 58 sweet corn inbred lines according to the sequence of Shrunken2 in B73. Total 7 635 bp sequence was obtained, which covered 20 exons (from exon 1 to exon 20), 17 introns (from intron 1 to intron 19, except intron 2 and intron 17) and 3'UTR region. From the analysis of gene polymorphism, a total of 105 polymorphism sites were identified, including 78 SNPs and 27 Indels. There were 58 polymorphism sites differences in shrunken2 sequences compared with that in Shrunken2 in B73, including 50 SNPs and 8 indels. Besides, 2 synonymous and 7 non-synonymous SNP mutations were found in the shrunken2 cording region. According to the shrunken2 gene sequence and coding region sequence, the inbred lines could be respectively divided into 14 and 4 haplotypes, Neutral evolution test showed that the shrunken2 gene was experienced both natural and artificial selection.
