摘  要:油质蛋白(OLE)是位于油体表面的一类膜结合蛋白,这类蛋白在油体形成过程中起重要作用。利用生物信息学手段在麻疯树基因组中共鉴定出6个JcOLE基因,并详细剖析这些基因的序列与进化特征。系统进化分析显示:JcOLE基因被分为5个类群,并在进化树末端发现了2对同源基因。序列分析表明:6个OLE基因具有3种基因结构类型,同时6个OLE蛋白具有3种跨膜结构与保守基序组成模式,共享了同一个典型OLE功能结构域。选择压力检测显示,JcOLE5和JcOLE6同源基因对受到负选择作用,但JcOLE1和JcOLE2同源基因对受到正选择作用,发生了适应性进化。EST数目与来源分析显示:除JcOLE4外,麻疯树OLE基因在胚中表达;JcOLE4在花中表达,而JcOLE3和JcOLE4在种子中表达。这些研究有助于进一步解析麻疯树OLE基因的生物学功能。Oleosin is a membrane protein localized on the surface of the oil body, which plays important roles in oil body formation and stability. Six JcOLE genes were identified using bioinformatics method in Jatropha curcas genome, sequence and evolutionary features of which were analyzed in detail. Phylogenetic analysis showed that all the JcOLE genes were divided into five groups, and that two pairs of homologous genes were found in the end of the phylogenetic tree. Sequence analysis indicated that three types of gene structures were identified among all the JcOLE genes. All the JcOLE proteins with the same functional domain had three types of transmembrane structure and conserved motif organization patterns. Selective pressure analysis demonstrated that JcOLE5 and JcOLE6 were under negative selection, but that JcOLE1 and JcOLE2 were under positive selection, suggesting adaptive evolution occurred after the divergence of JcOLE1 and JcOLE2. EST number and tissue source analysis demonstrated that, except for JcOLE4, five JcOLE genes were expressed in embryo. Additionally, JcOLE4 was expressed in flower, while JcOLE3 and JcOLE4 were expressed in seed. These results help to further investigate the biological function of the OLE family protein in Jatropha curcas.
