摘 要:目的探究邻苯二甲酸二异癸酯(di-iso-decyl phthalate,DIDP)经口暴露对过敏性皮炎的影响。方法用0.5%异硫氰酸荧光素(fluorescein isothiocyanate,FITC)构建过敏性皮炎模型,以褪黑素(melatonin,MT)为阻断剂,将雄性Balb/c小鼠随机分为7组:1生理盐水组;2200 mg/kg/d DIDP组;3FITC组;4FITC+2 mg/kg/d DIDP组;5FITC+20 mg/kg/d DIDP组;6 FITC+200 mg/kg/d DIDP组;7 FITC+200 mg/kg/d DIDP+MT组。1~7组每天用生理盐水或不同浓度的DIDP对小鼠进行灌胃,同时第7组小鼠还进行MT灌胃,连续21 d。第3~7组小鼠分别在第22、23、28 d进行致敏(0.5%FITC)。检测耳组织中的活性氧(reactive oxygen species,ROS)、还原型谷胱甘肽(reduced glutathione,GSH)、白介素-4(interleukin-4,IL-4)、干扰素-γ(interferon-γ,IFN-γ)和血清中的免疫球蛋白E(immunoglobulin E,Ig E)含量。结果高浓度的DIDP(200 mg/kg/d)可诱导耳组织产生氧化应激反应,并促进炎症因子的释放。MT处理能有效降低耳组织中的氧化应激水平,减轻过敏性皮炎的反应程度。结论 DIDP的长期暴露可使小鼠形成获得性过敏性体质,并通过增强氧化应激反应及下游分子事件,加重过敏原诱导的过敏性皮炎。Objective To explore the effect of di-iso-decyl phthalate(DIDP) on atopic dermatitis.Methods 0.5% of fluorescein isothiocyanate(FITC) was used to build a mouse model of allergic dermatitis,and melatonin(MT) was used as a blocking agent.Male Balb/c mice were randomly divided into seven groups:(1) saline group;(2) 200 mg/kg/d DIDPgroup;(3) FITC group;(4) FITC + 2 mg/kg/d DIDPgroup;(5) FITC + 20 mg/kg/d DIDPgroup;(6) FITC + 200 mg/kg/d DIDPgroup;(7) FITC +200 mg/kg/d DIDP + MT group.(1)-(7) groups were given a gavage of saline or three different concentrations of DIDPand grou P(7) was also given a gavage of MT once a day for continuous 21 days respectively.And subsequent three times of sensitization with 0.5% FITC on the 22 nd,23rd,and 28 thday.The concentrations of ROS,GSH,IL-4,IFN-γ in ear tissue and serum total Ig E were determined.Results High concentration of DIDP(200 mg/kg/d) could induce oxidative stress,and promote the release of inflammatory cytokines.MT could reduce the levels of oxidative stress and inflammatory cytokines significantly.Conclusion Longterm oral exposure to environmental toxins such as phthalates,and might endow an atopic predisposition in animals or humans.In addition,oxidative stress might play an important role in the aggravation effect.
