摘 要:目的:建立多重PCR技术鉴定食源性鼠伤寒沙门菌血清型的方法。方法:从60株菌株中随机选取4株食源性的沙门菌(分别命名为84号,95号,99号和06号),采用多重PCR法扩增其鞭毛抗原(H抗原)I相鞭毛素编码基因fliC和II相鞭毛素编码基因fljB,经测序比对鉴定各菌株的血清型。结果:多重PCR技术鉴定结果与生化鉴定法结果完全一致,84号菌株血清型模式为1,4:z4,12:e,h:1,2,其血清型为S.Saintpaul;95号血清型模式为1,4,12:i:1,2,其血清型为S.Typhimurium;99号菌株血清型模式为6,8:z4,z23:e,n,z15,其血清型为S.Chailey;06号菌株血清型模式为8,20:i:z6,不能确定其血清型。此外,测序结果还发现上述菌株存在新的变异位点。结论:本研究建立的多重PCR技术结果可靠,操作方便,适用于食品中沙门菌血清型的鉴定。Objective To establish a method to detect serotype of foodborne Salmonella typhimuriumwith a multiplex polymerase chain reaction (PCR) assay. Methods: Randomly select four strains which isolated from food (i. e., No. 84, No.95, No.99 and No.06) , and resequenced their genomic sequences of Flagellum I phase encoding genes (fliC) and Flagellum II phase encoding genes (fljB) with regard to the amplifiedsequences using a multiplex PCR assay. Each serotype of Salmonella typhimurium was identified by blastingResults: The serum model of four strains (84,95,99,06) were 1,4:z4,1 2 :e, h :1 ,2 ,1 ,4 ,1 2 :i :1 ,2 ,6 ,8 :z4,z23: e ,n,z15 and 8,20 : 1 : z6 , and the serotypes of which were S.Saintpaul, S.Typhimurium, S.Chailey respectively , but serotype of 06 can not be determined. In addition, several novel genetic variants were observed inthese strains. Conclusion: The multiplex PCR assay can be used to assess the serotype of foodborne Salmonella typhimurium, which has some practical value.
