摘  要:目的:探讨丹参酮ⅡA磺酸钠(STS)对阿霉素性心肌病大鼠心室肌闰盘的保护作用及机制。方法:30只Wister雄性大鼠分为对照组、阿霉素(ADR)组和STS+ADR组(均10只),对照组腹腔注射0.9%NaCl(10ml/kg),共3次(每周1次);ADR组腹腔注射ADR 2mg/kg,共注射3次(每周1次);STS+ADR组腹腔注射STS、ADR各2mg/kg,共注射3次(每周1次)。7周末各组大鼠行心脏彩超测左室舒张末径及左心室射血分数,各组大鼠处死后取左心室组织行透射电镜观察闰盘形态、结构,采用Western blotting技术检测各组心肌组织Cx40、Cx43及p-Cx43表达;ELISA法检测N-cad、DSG2的表达。结果:与ADR组相比较,STS+ADR组大鼠收缩与舒张功能明显改善(P<0.01),电镜下可观察STS+ADR组大鼠较ADR组大鼠心室肌闰盘结构破坏明显减轻,心室肌中Cx40、Cx43、p-Cx43、N-cad、DSG2表达明显增加(P<0.01)。结论:STS通过减轻ADR对闰盘结构的损坏,从而减少阿霉素心肌病心律失常的发生。Objective:To discuss the protective effect and mechanism of STS on ventricular muscle intercalated discs in rats with adriamycin-induced cardiomyopathy.Method:Sprague-Dawley male rats were randomly divided into control group(n=10),ADR group(n=10)and STS+ADR group(n=10).Intraperitoneal injection of 0.9%Nacl(10ml/kg)in control group three times(once per week).Intraperitoneal injected of ADR 2mg/kg in ADR group three times(once per week).STS+ADR group was intraperitoneal injected of STS 2mg/kg and ADR 2mg/kg three times(once per week).All the rats were sacrificed after measuring the Ultrasonic cardiogram(UCG)of LVIDd and LVEF on the 7th week.The morphology and structure of ventricular muscle intercalated discs was observed by using transmission electron microscope(TEM).The expression of Cx40,Cx43 and p-Cx43 were monitored by Western blotting and that of N-cad,DSG2 were monitored by ELISA.Result:Compared with ADR group,the cardiac systolic and diastolic function was improved significantly in STS+ADR group.Compared with ADR group,the destroy of ventricular muscle intercalated discs was relieved significantly and the expression of Cx40,Cx43,p-Cx43,N-cad and DSG2 was increased significantly in STS+ADR group.Conclusion:STS can reduce the incidence of arrhythmia in adriamycin induced cardiomyopathy,by reducing the ADR damage to the intercalated disc structure.
