摘 要:目的 建立同时测定紫锥菊维生素C胶囊中菊苣酸和维生素C含量的HPLC方法。方法 采用L9(34)正交试验优化提取条件。采用Hedera ODS-2 C18色谱柱(4.6mm×250mm,5μm),柱温35℃,流动相甲醇-乙腈(1∶1)-0.1%磷酸梯度洗脱,流速1.0m L·min-1,检测波长246nm。结果 维生素C和菊苣酸质量浓度分别在5~100μg·m L-1(r=0.9996)和10~200μg·m L-1(r=0.9996)范围内呈良好线性关系,平均加样回收率(n=9)分别为98.60%(RSD=2.53%)和102.08%(RSD=2.14%)。结论 本方法操作简便,准确度、精密度、稳定性良好,可用于紫锥菊维生素C胶囊的质量控制。OBJECTIVE To establish a method for simultaneous determination of cichoric acid and vitamin C in Echinacea and vitamin C capsules by HPLC. METHODS The L9 (3^4) orthogonal design was carried out in order to optimize the extraction method. The HPLC analysis was performed on a Hedera ODS-2 C18 column(4.6mm× 250mm, 5μm) at 35℃ ,with 0. 1% phosphoric acid and methanol-acetonitrile (1:1 ) as the mobile phase by gradient elution at a flow rate of 1.0mL · min-1. The detection wavelength was 246nm. RESULTS The good linearity of vitamin C and cichoric acid was achieved in the range of 5 - 100μg · mL-1 ( r = 0. 9996 ) and 10 - 200μg· mL-1 ( r = 0. 9996) ;the average recoveries (n =9) were 98.60% (RSD =2.53% ) and 102.08% (RSD =2. 14% )respectively. CONCLUSION The method is simple and proved to be accurate, reproducible and stable. It could be used to control the quality of Echinacea and vitamin C capsules.
