超高效液相色谱法测定皮革中多胺物质的研究

摘  要:皮革中氨基酸脱羧基可产生多胺类物质,毛皮鞣剂助染性聚酰胺多胺酚醛树脂,分解也可产生多胺类物质,多胺类物质具有致癌致畸性,目前国内外没有针对皮革材料建立多胺类物质的检测方法。研究通过振荡提取,超低温离心法净化提取液,柱前多胺与丹磺酰氯衍生,产物在254 nm荧光区处有最大吸收,建立了高效液相色谱检测皮革中多胺物质的方法。在优化的色谱条件下,检测低限在0.15 mg/L^0.20 mg/L,方法的回收率高于94.6%。方法被成功应用于测定皮革中的多胺物质。Leather amino acid decarboxylation can produce polyamine. The decomposition of fur tanning agent assisting dyeing of polyamide polyamine phenolic resin can also produce amines and polyamine material with carcinogenic teratogenic. At present, there is no material for leather established detection method of polyamine. It is by shaking extraction method, pre-column derivatization of polyamines with dansyl chloride and products at 254nm fluorescence region withe the maximum absorption. A simple method was followed by performance liquid chromatography which has been developed for the determination of polyamine in leather samples. Under the optimized conditions, the detection limit of the method was from 0.15 mg/L to 0.20 mg/L, and the recovery rate was higher than 94.6%. It could be applied for the determination of polyamine in leather.
