摘 要:为了解广东地区口蹄疫病毒(FMDV)遗传进化情况,本研究从广东采集5份疑似FMDV感染病料,对样品进行RT-PCR扩增,经克隆测序后,对FMDVs基因序列比对和遗传进化分析。鉴定结果显示,这些病料样品中2份为O型FMDV感染,3份为A型FMDV感染。2个FMDV O型样品中VP1基因的全长均为639 bp,编码213个氨基酸,3A基因的全长均为459 bp,编码153个氨基酸;3个FMDV A型样品中VP1基因的全长均为636 bp,编码212个氨基酸。3A基因的全长均为459 bp,编码153个氨基酸;与国内外分离的O/A型病毒株VP1基因核苷酸序列相比较发现,2个O型病毒VP1基因与O/BY/CHA/2010株核苷酸序列相似性最高(95.0%),均属于耿马谱系FMDV(属ESA基因型);3个A型病毒VP1基因与A/GDMM/CHA/2013株核苷酸序列相似性最高(99.2%),均属于Asia型FMDV。本研究通过对FMDV遗传进化分析为我国FMDV流行病学的研究提供了基础数据,同时对临床上不同型疫苗的选择提供依据。In order to analysis the genetic evolution of foot-and-mouth disease virus (FMDV) in Guangdong region, five clinical samples from suspected FMDV infected pigs were collected and detected for FMDV by RT-PCR, and 5 positive samples of FMDVs were detected from the clinical samples, of which 2 FMDVs were O type and 3 were A type. The ORF of VP1 gene in two O type FMDVs were 639 nt encoding 153 amino acids, and the 3A gene were 459 nt encoding 153 amino acids; the VP1 and 3A genes of the three A type FMDVs were 636 and 459 nt encoding 212 and 153 amino acids, respectively. Compared the VP1 gene with reference strains, the two O type FMDVs had the highest similarities (95%) with O/BY/CHA 2010 strain, and belonged to the social spectrum type (ESA genotype). In addition, the 3 A type FMDVs shared 99.2% nucleotide similarity with A/GDMM/CHA/2013 strain, belonged to Asia type. These data provide background information for efficient control the disease via vaccine selections against different types of FMDVs infections.
