黄淮白山羊冠状动脉和心肌组织结构及CX-43蛋白的分布观察

摘　　要：为观察黄淮白山羊冠状动脉、心肌组织结构及其CX-43蛋白的分布特点,将10只健康成年黄淮白山羊经麻醉后颈动脉放血致死,取其心制作标本。一部分通过冠状动脉血管铸型标本制备,心肌及冠状动脉标本常规石蜡切片,经MASSON三色、VVG和免疫组织化学染色后显微观察并摄影;另一部分冠状动脉标本通过戊二醛磷酸盐缓冲液固定后,于60℃300 g/L的KOH溶液中浸泡、0.1 mol/L磷酸盐缓冲液冲洗、37℃胶原纤维酶Ⅱ溶液处理6 h后通过20 g/L鞣酸浸泡2.5 h,10 g/L四氧化锇浸泡、梯度乙醇脱水,喷镀氯化金,扫描电子显微镜观察并摄影。结果表明,黄淮白山羊冠状动脉主要形成左冠状动脉旋支、前降支、左前降支对角支及窦房结支;右冠状动脉可形成右冠状动脉旋支,前、后降支和窦房结支等重要分支,黄淮白山羊冠状动脉属于左优势型。弹性冠状动脉管壁中弹性纤维捆扎成束,部分弹性纤维呈螺旋状,肌性冠状动脉管壁平滑肌丰富,少见或偶见弹性纤维。隔缘肉柱心内膜下方分布有浦肯野细胞束,小动脉位于其近中央分布;乳头肌细胞排列整齐,细胞间有少量胶原纤维,浦肯野细胞束沿乳头肌中血管边缘分布;心耳心肌细胞间的胶原纤维明显多于室中隔心肌细胞间的胶原纤维含量;CX-43蛋白主要分布于隔缘肉柱、乳头肌、心房和室中隔的心肌细胞连接处和浦肯野细胞胞膜。To observe characters of coronary artery, myocardial tissue structure and CX-43 protein distribution in Huanghuai white goat,ten healthy adult Huanghuai white goats were died by carotid artery bleeding to death after anesthesia, taking its heart specimens. A part of the coronary artery was prepared into vascular cast specimens, and myocardium and coronary arteries were made into paraffin section, by MASSON, VVG and immunohistochemical staining, microscopic observation and photography. Another part of the coronary artery specimens were fixed in glutaraldehyde phosphate buffer, at 60℃ in 300 g/L KOH solution soak, 0. i mol/L phosphate buffer flushing, after 6 h treatment of 37℃ coilagen enzymell solution, by 20 g/L tannic acid soaking for 2.5 h, then 10 g/L osmium tetroxide soaking,gradient alcohol dehydration, and plating gold chloride and scanning electron microscope observation and photography. The results showed that the main left coronary artery embranchments in Huanghuai white goats had circumflex branch of left coronary artery, left anterior descending artery, diagonal branch of left anterior descending artery and branch to sinoatrial node. The main right coronary artery em- branchments in Huanghuai white goats had a circumflex branch of right coronary artery, right anterior descending artery, right posterior descending artery and branch to sinoatrial node. The coronary artery in Huanghuai goat belongs to the left advantage type. Elastic fibers were strapped into a bundle in the artery wall; a part of the elastic fibers presented a spiral shape, and the artery wall had rich smooth muscle in muscle muscular, rare or occasional seen elastic fibers. There were purkinje fibers bundles at the bottom of septomarginal trabecular tunica, a small artery located in the close central of it. Papillary muscle cells arranged trimly, and had a small amount of collagen fibers between them.
