摘 要:目的:研究千佛菌对1型糖尿病模型小鼠细胞因子基因异常表达的调节作用及血糖水平、小鼠行为变化的影响,以探讨千佛菌对1型糖尿病小鼠模型的双向免疫调节作用。方法:观察千佛菌大、小剂量组、模型对照组与正常对照组小鼠行为变化;血糖的水平;淋巴细胞IL-2m RNA、IL-10m RNA的表达水平。结果:模型对照组小鼠表现为"三多一少"的症状:饮食多,饮水多,尿量增多,体型瘦弱。千佛菌大、小剂量组症状较模型对照组有所减轻,而正常对照组未见异常。与正常对照组比较,模型对照组血糖水平、IL-2m RNA表达水平明显增高(P<0.01),而IL-10m RNA表达水平明显降低(P<0.01);与模型对照组比较,千佛菌大、小剂量组的血糖水平、IL-2m RNA表达水平均明显降低(P<0.01、P<0.05),IL-10m RNA表达水平均明显增高(P<0.01、P<0.05);而千佛菌大、小剂量组的血糖水平、IL-2m RNA及IL-10m RNA表达水平的差异无统计学意义(P>0.05)。结论:千佛菌具有降低1型糖尿病模型小鼠血糖的作用,能下调IL-2m RNA的表达,上调IL-10m RNA的表达,对1型糖尿病模型小鼠脾脏淋巴细胞Thl/Th2细胞因子的平衡具有调节作用。Objective: In order to investigate the two- way adjustment of Grifola frondosa against mice with type 1diabetic mellitus( T1DM). Methods: The behavior of all mice was observed. The glucose level in blood was tested by Blood Glucose Meter. The RNA was extracted in the spleen and the IL-2m RNA and IL-10 m RNA expression of Th1 / Th2 cytokines were detected by Real-time PCR. Results: The model of T1 DM showed the symptoms of polydipsia,polyuria,poly-phagia and weight loss. The symptoms of the mice in the therapy group were alleviated. Compared with the controlled group,the glucose level in blood and the expression of IL-2m RNA all increased obviously( P0. 01),but the expression of IL-10 m RNA decreased significantly in model group( P0. 01); compared with the model group,the glucose level in blood and the expression of IL- 2m RNA all decreased significantly( P0. 01,P0. 05),but the expression of IL-10 m RNA all increased obviously in therapy group 1and group 2( P0. 01,P0. 05). However,there was no significant difference in the glucose level in blood and the expression of IL- 2m RNA and IL-10 m RNA between the therapy group 1and group 2( P0. 05). Conclusion: Grifola frondosa is beneficial to lower the glucose level in blood in mice with T1 DM. At the same time,it can also decrease significantly the expression of IL-2m RNA and enhance the expression of IL-10 m RNA. Grifola frondosa plays a role in regulating the balance of Th1 / Th2 cytokines by the two-way adjustment.
