摘  要:目的探讨富硒灵芝多糖对非酒精性脂肪肝病大鼠SREBP-1c及ACCα表达的影响。方法 120只SD大鼠随机分为正常组、高脂饮食组、多烯磷脂酰胆碱组、富硒灵芝多糖低、中和高剂量干预组等6组,每组20只,分别于治疗4和8周后处死大鼠各半,测血清中谷草转氨酶(AST)、谷丙转氨酶(ALT)、总胆固醇(CHOL)和三酰甘油(TG);用Western blot和RT-PCR检测肝脏组织中SREBP-1c及ACCα蛋白和mRNA表达;其余肝脏行病理学检测。结果富硒灵芝多糖可显著减少肝脏组织中的脂肪颗粒,并降低血清中AST、ALT、CHOL和TG含量,中、高剂量的灵芝多糖处理可显著降低肝脏中SREBP-1c、ACCαmRNA和蛋白的表达(P<0.05)。结论富硒灵芝多糖可缓解高脂饮食诱导的非酒精性脂肪肝大鼠症状。Objective To investigate the expression of ACCα and SREBP-1 c of selenium-enriched Ganoderma luci- dum polysaccharides on fatty liver rats model. Methods One hundred and twenty SD rats were randomly divided into control, high-fat diet, polyene phosphatidylcholine, Se-G. lucidum polysaccharides low, medium and high group, after 4 and 8 weeks of treatment the rats were sacrificed, measuring the AST, ALT, CHOL and TG in blood serum, detect SREBP-1c, ACCα mRNA and protein expression by Western blot and PCR respectively. Results With the increase of treatment dose and incubation time of Se-G. luciclum polysaccharides, fatty infiltration of the liver was significantly reduced, fat particles significantly decreased; liver function and blood lipids profile signifi- cantly improved; SREBP-1 c, ACCα mRNA and protein expression was significantly decreased. Conclusions Se- G. lucidum polysaccharides can ameliorate the symptoms of non-alcoholic fatty liver disease in rats.
