摘 要:目的探索小鼠卵巢生殖干细胞(OSCs)卵巢内移植是否可以改善化疗诱导的卵巢损伤。方法采用腹腔注射环磷酰胺和白消安建立化疗诱导的小鼠卵巢损伤模型,并随机分为模型组及移植组,同龄小鼠作为正常对照组。移植组在建模1周后,双侧卵巢分别移植OSCs。移植后观察小鼠动情周期;检测血清雌二醇(E2)和卵泡刺激素(FSH)浓度;测量卵巢重量及直径;卵巢组织切片观察卵巢结构和卵泡数目。结果 90%的模型组小鼠动情周期紊乱,70%移植组小鼠恢复正常的动情周期。移植组E2平均浓度(55.8±6.342)pg/ml高于模型组(34.1±4.643)pg/ml,差异有统计学意义(t=6.758,P<0.05),而FSH浓度(28.3±1.433)ng/ml低于模型组(64.9±5.553)ng/ml,差异有统计学意义(t=-15.631,P<0.05)。模型组卵巢呈萎缩状态,而移植组卵巢得到明显改善,形态、大小和正常对照组相似。移植组卵巢内各级卵泡数目均高于模型组,差异有统计学意义(t=11.851,P<0.05),但仍低于正常对照组,差异亦有统计学意义(t=6.505,P<0.05)。结论卵巢生殖干细胞卵巢内移植可以部分地修复化疗导致的卵巢损伤,从而改善卵巢功能。Objective To explore whether ovarian reproductive stem cell transplantation of mice can improve chemotherapy-induced o- varian damage. Methods Intraperitoneal injection of cyclophosphamide and busulfan was used to establish mouse mode of ovarian damage induced by chemotherapy, then the mice were randomly divided into model group and transplantation group, the mice of the same age were selected as normal control group. After establishing the model for one week, ovarian reproductive stem cell transplantation of bilateral ovaries was conducted among the mice in transplantation group. After transplantation, estrous cycle of mice was observed ; the concentrations of serum estradiol and follicle stimulating hormone (FSH) were detected, the weight and diameter of ovary were measured, and ovarian tissue sections were examined to observe Ovarian structure and the number of follicles. Results Estrous cycle disorder was found among 90% of the mice in model group, estrous cycle recovered to normal among 70% of the mice in transplantation group. The average serum concentration of estradiol in transplantation group was (55.8±6. 342) pg/ml, which was statistically significantly higher than that in model group [ (34. 1 ± 4. 643) pg/ml] (t=6. 758, P〈0. 05). The average serum concentration of FSH in transplantation group was (28.3±1. 433) ng/ml, which was statistically significantly lower than that in model group [ (64. 9±5. 553 ) ng/ml ] (t =- 15.631, P〈0. 05 ). The ovaries of mice in mode] group were atrophic, but the ovaries of mice in transplantation group were improved significantly, which were similar tO ovaries of mice in normal control group in aspects of morphology and size. The numbers of follicles at different levels in ovaries of mice in transplantation group were statistically significantly higher than those in model group (t = 11.851, P〈0. 05 ), but they were still statistically significantly lower than those in normal control group ( t = 6. 505, P 〈 0. 05 ) .
