青蒿素抑制心肌缺血再灌注中的细胞凋亡

摘　　要：目的:探讨青蒿素对心肌缺血再灌注(I/R)损伤的影响以及其可能的机制。方法:将30只雄性SD大鼠随机分为假手术(SO)组、缺血再灌注(I/R,缺血30 min,再灌注4 h)组和青蒿素组(A+I/R,青蒿素25 mg·kg-1,每天3次,造模前1 d灌胃)组,每组10只。检测各组肌酸激酶(CK)、乳酸脱氢酶(LDH)、肌钙蛋白I(c Tn I)、丙二醛(MDA)、超氧化物歧化酶(SOD)、高迁移率族蛋白1(HMGB1)、B淋巴细胞瘤2(Bcl-2)、Bcl-2相关X蛋白(Bax)和活化含半胱氨酸的天冬氨酸蛋白水解酶(cleaved-caspase-3),并进行比较分析。结果:与SO组比较,I/R组心肌损伤标志物(CK、LDH和c Tn I)、氧化应激水平(MDA和SOD)、HMGB1和凋亡蛋白(Bcl-2、Bax和cleaved-caspase-3)的表达增加。与l/R组比较,青蒿素组心肌损伤标志物、氧化应激水平、HMGB1和凋亡蛋白的表达显著降低。结论:青蒿素可能通过抑制HMGB1和氧化应激,抑制I/R诱导的心肌细胞凋亡,减轻心肌I/R损伤。Objective： To investigate the effects and the possible mechanism of artemisinin on myocardial ischemia and reperfusion（ I / R） injury. Methods： Thirty male SD rats were divided into three groups： sham operate（ SO） group,ischemia and reperfusion（ I / R） group（ ischemia 30 min and reperfusion 4 h） and artemisinin treatment（ A ＋ I / R） group（ arteannuin 25 mg · kg（-1）,ig,Tid,one day before the sugery）,10 for each group.Serum creatine（ CK）,lactate dehydrogenase（ LDH）,cardiac troponin I（ c Tn I）,malondialdehyde（ MDA）,superoxide dismutase（ SOD）,high mobility group box protein 1（ HMGB1）,B cell lymphoma / lewkmia-2（ Bcl-2）,Bcl-2 Assaciated X protein（ Bax） and cleaved-caspase-3 were detected and compared among the 3 groups.Results： Compared with SO group,cardiac injury markers（ serum CK,LDH and CK）,oxidative stress（ MDA and SOD）,HMGB1 and apoptosis level（ Bcl-2,Bax and cleaved-caspase-3） expression significantly increased.Compared with I / R group,cardiac injury markers,oxidative stress,HMGB1 expression and suppressed apoptosis were significantly decreased in artemisinin group. Conclusions： Artemisinin might inhibite apoptosis in myocardial I / R injure via suppressing HMGB1 expression and oxidative stress.
