MK-801对阿尔茨海默病模型大鼠学习记忆能力及Aβ表达的影响

摘　　要：目的:研究地卓西平(MK-801)对阿尔茨海默病(AD)模型大鼠学习记忆能力及神经元固缩、β-淀粉样多肽(Aβ)表达的影响。方法:将32只雄性SD大鼠随机分为AD模型+MK-801、AD模型+生理盐水、假手术+MK-801、假手术+生理盐水4组。向大鼠侧脑室注射链脲佐菌素(STZ)制备AD模型,侧脑室注射柠檬酸缓冲液制备假手术模型。造模后10 d,4组分别腹腔注射MK-801或生理盐水,连续30 d。在造模后10 d和给药后30 d行水迷宫测试。最后将脑组织制作石蜡切片,HE染色观察神经元固缩率,免疫组织化学染色观察神经元Aβ阳性率。结果:给药后30日,AD模型+MK-801组比AD模型+生理盐水组大鼠潜伏期缩短,跨台次数增加,神经元固缩率和Aβ阳性率减少,差异有统计学意义(P<0.05);比较AD模型两组与假手术两组的差异:腹腔注射MK-801比生理盐水潜伏期差异缩短,跨台次数差异增加,细胞固缩率和Aβ阳性率差异减少,此变化有统计学意义(P<0.05)。结论:腹腔注射MK-801能改善AD模型大鼠的学习记忆功能障碍,抑制神经元固缩、Aβ沉积。Objective： To study the effect of dizocilpine（ MK-801） on the learning,memory function and the neuronal pycnosis,expression of Aβ in rats of Alzheimer＇s disease（ AD） model. Methods： Thirty-two male SD rats were divided into model ＋ MK-801,sham operated ＋ MK-801,sham operated ＋ saline and model ＋ saline groups,randomly. The models of Alzheimer＇s disease were created by intracerebroventricular injection of streptozotocin（ STZ）. MK-801 or saline were administered to the 4 groups. All groups were tested on Morris water maze on day 10 and day 30. The pycnosis rate was observed by HE staining and the Aβ positive neuron rate by immunohistochemistry staining at last. Results： Thirty days later,AD model rats who were given MK-801 got shorter latencies,less passing through platform times and pyknosis rate and Aβ positive neuron rate reduced,compared to AD model rats who were given saline,this change was statistically significant（ P〈0. 05）. Comparing the difference between the AD model group and sham operated group： intraperitoneal injection of MK-801 reduced latencies gap,increased passing through platform times gap and reduced pyknosis rate and Aβ positive neuron rate gap compared to saline,this change was statistically significant（ P〈0. 05）. Conclusion： Injection of MK-801 can improve the learning and memory capabilities of AD model rats,reduce neuronal pyknosis and deposition of Aβ.
