摘  要:目的探讨具有同源受体CCR4的趋化因子CCL17和CCL22对CD4和CD8 T细胞亚群的趋化能力。方法首先利用流式细胞术检测健康人外周血中CCR4^+T细胞的比例分布,接着用Mo Flo分选流式细胞仪分选出健康人外周血中的CD4细胞和CD8细胞。利用CCL17和CCL22趋化因子蛋白分别对CD4及CD8细胞进行细胞趋化实验,计算趋化指数。利用流式细胞术检测CCL17和CCL22分别作用CD4及CD8细胞后CCR4受体内化情况,计算受体内化效率。结果外周血PBMC中,CD4细胞CCR4分子的表达水平要高于CD8细胞(P<0.05)。在相同趋化时间内,CCL22对CD4和CD8 T细胞亚群的趋化能力强于CCL17(P<0.05),CCL17和CCL22趋化CD4细胞通过Transwell板的细胞数均多于CD8细胞(P<0.05),但CCL17对2种T细胞亚群趋化能力的差异性要小于CCL22。CCR4受体内化实验证实,相同时间下CCL22比CCL17更容易促使CCR4受体发生内化(P<0.05),对于CD4和CD8 T细胞亚群,CCL22作用后受体CCR4内化程度无显著性差别(P>0.05),而CCL17对CD8细胞的内化作用要强于CD4细胞(P<0.05)。结论 CCL22对CD4和CD8细胞的趋化能力均强于CCL17,而促进受体内化方面,CCL17对CD8细胞的作用更强,CCL22对这2种细胞则无显著差别。This study designed to investigate the chemotaxis of CCL17 and CCL22 with homologouschemo+kine receptor CCR4 in CD4 and CD8 cells. Firstly, flow cytometry was used to detect the proportion ofCCR4 T cells in the peripheral blood of healthy people, and then the CD4 cells and CD8 cells in peripheral bloodof healthy people were selected by Mo Flo sorting flow cytometry. Chemotaxis experiments were done on CD4 andCD8 cells with CCL17 and CCL22 chemokine protein. Finally, CCR4 receptor internalization in CD4 and CD8 cells were detected after CCL17 and CCL22 chemotaxis. Flow analysis showed that the expression level of CCR4 in CD4 cells was significantly higher than that in CD8 cells(P〈0.05). During the same time, the number of CD4 cells attracted through Transwell by CCL17 and CCL22 were more than that of CD8 cells, while the overallchemotaxis of CCL22 was stronger than that of CCL17(P〈0.05). CCR4 receptor internalization experimentsconfirmed that CCL22 was more likely to cause CCR4 receptor internalization, as compared with CCL17(P〈0.05). The internalization of receptor CCR4 showed indifference on CD4 and CD8 cell after CCL22 treatment(P〉0.05), but after CCL17 treatment, the internalization of receptor CCR4 on CD8 cells was stronger than that onCD4 cells(P〈0.05), suggesting that the interaction between CCL17 and CD8 cells was stronger. Taken together,the chemotaxis of CCL22 is relatively stronger than CCL17 on CD4 cells and CD8 cells. In terms of receptor internalization, CCL17 has a stronger effect on CD8 cells than that on CD4 cells, while CCL22 has nodifference.
