摘  要:目的探讨金花菜对营养性肥胖大鼠体重及血脂的影响及可能作用机制。方法采用高脂饲料喂养9周建立营养性肥胖大鼠模型,将50只造模成功大鼠随机分为模型对照组、阳性对照组和金花菜高、中、低剂量组,每组10只,另设正常大鼠10只为空白对照组。金花菜高、中、低剂量组分别给予金花菜混悬液2、1、0.5 g/(kg·d)灌胃,阳性对照组给予奥利司他胶囊35 mg/(kg·d)灌胃,空白对照组、模型对照组给予同体积(15 ml/kg)的蒸馏水灌胃,连续6周。于给药前后测量大鼠体重计算总增重,给药后检测血清甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)水平和肝脏卵磷脂胆固醇脂酰转移酶(LCAT)、胆固醇7α-羟化酶(CYP7A1)、过氧化物增殖激活物受体α(PPARα)mRNA和蛋白表达。结果与模型对照组比较,各给药组大鼠给药后体重及总增重明显下降,金花菜高剂量组和阳性对照组TC、TG和LDL-C明显降低,HDL-C显著升高(P<0.05或P<0.01)。治疗后各组肝脏组织PPARα和CYP7A1 mRNA表达差异无统计学意义(P>0.05)。与模型对照组比较,金花菜高剂量组PPARα蛋白表达升高,而阳性对照组PPARα、LCAT的蛋白表达显著降低(P<0.05或P<0.01)。结论金花菜对营养性肥胖大鼠有较好的减肥降脂作用,其机制可能与激活PPARα蛋白表达有关。Objective To explore the impact of medicago polymorpha on weight and blood fat of lipid in alimentary obesity rats and the possible mechanism. Methods SD rats were fed with high fat diet for 9 weeks to establish alimentary obesity models of rats. Fifty successfully modeled rats were randomized into model control group, positive control group, high, middle and low dose medicated group, 10 rats in each group. Ten normal and healthy rats were set as blank control group. The high, middle and low dose medicated groups were given medicago polymorpha suspension 2, 1,0. 5 g/( kg · d) respectively by gavage. The positive control group was given orlistat capsules 35 mg/( kg · d) by gavage. The blank control group and the model control group were given equivoluminal (15 ml/kg) distilled water by gavage for 6 weeks. Body weights were measured before and after treatment, and total weight increment was calculated. Levels of serum triglyceride (TG), low density lipoprotein cholesterol (LDL-C), total cholesterol (TC) and high density lipoprotein cholesterol ( HDL-C ), as well as the expressions of liver lecithin cholesterol acyl transferase (LCAT), cholesterol 7a-hydroxylase (CYP7A1), peroxide proliferation activated receptom (PPARot) mRNA and proteins were determined. Results Compared with the model control group, body weights and total weight in- crement after administration in each drug administration group decreased. TC, TG and LDL-C in the high dose medi- cated group and the positive control group decreased, while HDL-C increased ( P 〈 0.05 or P 〈 0. 01 ). After treat- ment, the difference of the expressions of liver tissue PPARa and CYP7A1 mRNA in each group was not significant (P 〉 0. 05). Compared with the model control group, the expression of PPARα protein in the high dose medicated group increased. While the expressions of PPARα and LCAT proteins in the positive control group decreased (P 〈 0. 05 or P 〈 0. 01 ). Conclusion medicago polymorpha might have
