摘　　要：目的探讨宫宁颗粒诱导IUD避孕大鼠血管平滑肌细胞(VSMC)凋亡的影响。方法将50只已生育SD雌性大鼠随机分为宫宁颗粒组、消炎痛组、模型组、假手术组、空白对照组,应用光镜观察大鼠子宫组织病理学变化;免疫组化法检测大鼠子宫VSMC中Bcl-2、Bax、Fas和FasL蛋白表达。结果置入IUD后大鼠子宫内膜及血管结构功能发生异常,与模型组比较,宫宁颗粒能显著降低子宫VSMC中Bcl-2蛋白表达,增加Bax、Fas、FasL蛋白表达,并降低Bcl-2/Bax值(P<0.01),且效果明显优于消炎痛组(P<0.01)。结论宫宁颗粒可减轻大鼠子宫内膜形态学损伤,消除子宫血管构筑的异常,其机制可能是通过升高Bax、Fas、FasL蛋白表达,降低Bcl-2蛋白表达,诱导子宫VSMC凋亡,进而达到防治置环后异常子宫出血的目的。Objective To explore the effect of GongNing Granules on apoptosis of vascular smooth muscle cells （VSMC） from rats with the intrauterine device. Methods 50 SD female infertile rats were randomly divided into blank control group, model group, sham operation group,indomethacin group and GongNing granule group. Light microscopy was applied to each group of rats for observing the uterine tissue pathological changes ; immunohistochemical method was applied to each group of rats to detect the protein expression of Bcl - 2, Bax, Fas, FasL in the uterine VSMC. Results Inserting IUD can result in abnormal structure and function of the rats＂ endometrium and vascular. Compared with the model group, the protein expression of Bcl - 2 and the ratio of Bel -2 to Bax in GongNing granule group was significantly decreased, but the protein expression of Bax, Fas, FasL were signifi- cantly increased （P 〈 0.01 ）. The effect was obviously better than those in indomethacin group （P 〈 0.01 ）. Conclusion GongNing granules could lighten the endometrial morphology damage and eliminate uterine abnormal vessel architecture of the rats, the mechanism might promote the uterine VSMC＇s apoptosis by increasing the protein expression of bcl -2 and decreasing the protein expression of Bax, Fas, FasL in the uterine VSMC, further to achieve the object of preventing and treatment of abnormal bleeding after inserting IUD.
