摘 要:目的通过实验性肿瘤模型对莪术、白术、连翘三种中药挥发油处方比例进行优选,并对处方抗肿瘤效果进行评价。方法采用S180瘤株肿瘤模型,利用L9(3)4正交试验设计,以肿瘤抑制率为指标,对白术、莪术、连翘挥发油的剂量配比进行优选。对优选处方的抑瘤效果进行评价,以环磷酰胺为阳性对照药组,观察肿瘤抑制率、脾指数和胸腺指数。结果三种挥发油抗肿瘤的最佳比例为白术油500mg/kg,莪术油100mg/kg,连翘油25mg/kg。对优选出的挥发油剂量进行了抑瘤效果的评价,低剂量、中剂量、高剂量的肿瘤抑制率分别为29.08%、41.15%、58.20%。挥发油的各剂量组和环磷酰胺组的脾指数均小于空白组,与空白组相比均有显著性差异(P<0.05)。挥发油的中剂量组、高剂量组和环磷酰胺组的胸腺指数均低于空白组,与空白组相比有显著性差异(P<0.05)。结论正交实验优选确定了三种挥发油配伍剂量,三者合用具有一定的抗肿瘤作用。Objective To optimize the three volatile oil of Rhizoma Zedoariae, Forsythia and Atractylodes Rhizome scale by the medium of experimental tumor model, ascertain prescription dosage and appraise the antitumor effect. Methods Taking the tumor inhibition ratio as experimaent indices, using orthogonal test to optimise the dosage of Atractylodes, Zedoary, Forsythia volatile oil and appraise antitumor effect of the prescription. The tumor vaccine strains of mice were randomly divided into drug cyclophospha- mide positive control group, blank control group, model group, as well as large, medium and small dose group, killed the mice after 7 days'administration, peeled tumor, spleen and thymus, calculated tumor inhibition rate, spleen index and thymus index. Results The best ratio three kinds of volatile oil were Atractylodes oil 500mg/kg, Zedoary oil 100mg/kg, Forsythia oil 25mg/kg. Appraising the antitumor effect of optimized volatile oil: the inhibition ratio of large, middle, smallness dosage is 29.08%, 41.15% ,58.20%, proceeding statistical treatment. Each volatile oil administration group has significant difference compared with model set. (P 〈 0.05) Furthermore, the inhibition ratio of each group is higher than the simple oil. The spleen indices of each dosage group of volatile oil and CTX group is less than blank group, in comparison with blank group equal have got significant difference. (P 〈 0.05) The thymus indices of middle dosage group, high dose group and CTX group is less than blank group, in comparison with blank group have got significant difference(P 〈 0.05 ). Conclusion The optimized scale antitumor effect is obvi- ously, and the combination of volatile oil have the definite antitumor sense.
