摘  要:目的探讨干细胞因子(stem cell factor,SCF)对活动低下逼尿肌中Cajal间质细胞(interstitial cells of Cajal,ICCs)数及逼尿肌收缩力的影响。方法将SD大鼠分为3组:对照组、逼尿肌活动低下(detrusor underactive,DU)组以及DU+SCF组。采用ELISA检测血清SCF水平,免疫荧光组织化学法检测ICCs细胞数目,RT-PCR以及Western blot检测c-kit以及SCF的组织水平表达;体外肌条实验检测逼尿肌收缩能力。结果活动低下逼尿肌组织中ICCs数显著减少(P<0.05),SCF及c-kit表达显著降低(P<0.05),逼尿肌收缩频率及收缩幅度显著下降(P<0.05);外源性SCF可以显著增加活动低下逼尿肌组织中ICCs数(P<0.05),增加SCF及c-kit的表达(P<0.05),显著提升逼尿肌收缩幅度和收缩频率(P<0.05)。结论外源性SCF可以影响活动低下逼尿肌组织中ICCs数,进而改变逼尿肌收缩能力。Objective To determine the effect of stem cell factor (SCF) on the number of interstitial cells of Cajal (ICCs) and the contraction of detrusor in the rats with underactive bladder. Methods A total of 54 SD rats were randomly divided into 3 groups : control group, detrusor underaetivity group ( DU, inflicted with bladder outlet obstruction), and DU + SCF group [ intra-peritoneal injection of 0.2 μg/(kg · d) SCF], with 18 animals in each group. After the rats were sacrificed in 2 weeks later, their blood and bladder samples were harvested. The number of ICCs was analyzed by immunohistochemical and immunofluoreseent staining. The serum SCF levels were detected by ELISA. The expression levels of c-kit and SCF in the detrusor tissues were detected by RT-PCR and Western blotting. Change of detrusor spontaneous contraction was tested with detrusor strips in vitro. Results The number of ICCs was decreased significantly, the serum SCF level was also reduced significantly, so were the expression of c-kit and SCF at mRNA and protein levels, and the contraction of detrusor was decreased significantly in the DU group compared to the control group ( all P 〈 0.05 ). However, exogenous SCF resulted in the increase of ICCs number, enhancement of c-kit and SCF expression levels, and significant improvement of the contraction amplitude and frequence of detrusor ( all P 〈 0. 05). Conclusion Exogenous SCF improves the number of ICCs and the contraction of detrusor in DU rats.
