HPLC法同时测定南大青叶中5种有效成分的含量

摘　　要：目的建立同时测定南大青叶中5种有效成分的高效液相色谱（HPLC）法。方法采用ZORBAX Extend-C18色谱柱（4.6 mm×250 mm,5μm）,流动相为乙腈（A）-水（B）,梯度洗脱,流速为1 m L/min,检测波长为225 nm,柱温为30℃。结果 2,4（1H,3H）-喹唑二酮、4（3H）-喹唑酮、2-甲基-4（3H）-喹唑啉酮、色胺酮、靛玉红进样量分别在10.86-217.2、16.21-324.2、11.34-226.8、29.85-597.0、160.5-3 210 ng范围内线性关系良好,加样回收率为98.5%-100.9%,方法学考察结果良好。结论建立的HPLC法简便快速、重复性好、结果准确可靠,可作为南大青叶药材质量标准控制的方法。Objective To establish the HPLC method for simultaneous determination of 5 active components in leaves of Baphicacanthus cusia（ Nees） Bremek.Methods A ZORBAX Extend-C18column（ 4.6 mm× 250 mm,5 μm） was used and eluted gradiently with the mobile phase of acetonitrile and water at a flow rate of1 m L/min.The detection wavelength was 225 nm and the column temperature was 30 ℃.Results 2, 4（ 1H,3H）-quinazolinedione,4（ 3H）-quinazolinone,2-methyl-4（ 3H）- quinazolinone,tryptamine ketone and indigo showed good linearities within the range of 10.86-217.2 ng,16.21-324.2 ng,11.34-226.8 ng,29.85-597.0 ng and 160.5- 3 210 ng, respectively.The recovery of them were among 98.5%- 100.9%.Conclusion The established HPLC method is simple, rapid, reproducible, accurate and reliable, which could be used to control the quality standard of leaves of B.cusia.
