摘 要:目的研究和比较两种鹿鞭水提物对顺铂(CDDP)诱导小鼠急性肾损伤保护作用,并探讨可能机制。方法 64只雄性小鼠随机均等分为空白对照组、CDDP模型组、新西兰马鹿鞭和梅花鹿鞭水提物高、中、低剂量(100μg/g、200μg/g、400μg/g)+CDDP保护组;各鹿鞭保护组连续灌胃7天,在第5天给药1 h后(除空白对照组)小鼠均腹腔注射CDDP(20μg/g)诱导急性肾损伤。造模48 h后,测定小鼠血清和肾组织中肾功能指标水平和观察HE染色肾脏组织病理学切片变化。结果与CDDP模型组比较,两种鹿鞭水提物各剂量保护组均可改善肾功能指标水平和肾损伤状况,梅花鹿鞭较新西兰马鹿鞭效果更佳,其中梅花鹿鞭水提物高剂量保护组效果最为显著:BUN、CRE和MDA显著降低(P<0.01),SOD和GSH显著升高(P<0.01);明显改善损伤肾脏中肾小管变性和坏死的现象,但未恢复正常水平。结论两种鹿鞭水提物均对顺铂诱导小鼠急性肾损伤有明显保护作用,可能与清除自由基和抗氧化应激作用有密切联系。Objective To investigate and compare the protective effect of two kinds of aqueous extractive of Penis et Testis Cervi against cisplatin(CDDP)-induced acute kidney injury in mice and discuss the possible mechanism. Methods 64 mice were evenly and randomly divided into the control group, CDDP model group, aqueous extract of penis cervi of Cervus nippon and Cervus elaphus with low-, mid- and high-dose (100,200,400 mg/kg respectively)groups, 8 mice in each group. The different groups of Penis et Testis Cervi were intragastrical administrated with corresponding aqueous extract for 7 days, the mice ( except the control group) were intraperitoneally injected with CDDP (20μg/g)to induce acute kidney injury on fifth day l h after administration. After modeling 48 h, the index level of renal function in serum and kidney tissue was measured and the pathological change was observed by HE staining. Results Compared with CDDP model group, the groups treated with two kinds of aqueous extract of Penis et Testis Cervi against with different dose were all improved the index level of renal function and renal damage, and the effect of penis cervi in Cervus nippon was better than Cervus elaphus. It showed the most significant effect in the group treated with penis cervi of Cervus nippon at high dose,which the contents of BUN, CRE and MDA significantly decreased( P 〈 0.01 )and the activity of SOD and GSH significantly increased ( P 〈 0.01 ). It obviously improved the degeneration and necrosis of renal tubule, but not recovered. Conclusion These two kinds of aqueous extract of Penis et Testis Cervi effectively protected mice from the acute kidney injury induced by cisplatin. It may be closely related to cleaning out radicals and anti-oxidative stress.
