摘 要:目的:观察补肾解毒方对^(60)Coγ射线辐射后小鼠TLR4(Toll like receptor-4)信号转导通路下游相关炎性因子表达的影响并探讨其作用机制。方法:将背景为C57BL/10J的小鼠(TLR4^(+/+))完全随机分为TLR4^(+/+)空白对照组、TLR4^(+/+)辐射模型组、TLR4^(+/+)中药组,同等背景下TLR4基因敲除小鼠(TLR4^(-/-))完全随机分为TLR4^(-/-)空白对照组、TLR4^(-/-)辐射模型组、TLR4^(-/-)中药组。除TLR4^(+/+)空白对照组、TLR4^(-/-)空白对照组外,其余四组小鼠均一次性接受^(60)Coγ射线6Gy剂量的全身照射,观察存活情况;记录各组小鼠辐射后第1、14、30天脾脏IL-6、IL-10、IL-17、IL-18、IFN-γ、TGF-β1的数值。结果:TLR4^(+/+)辐射模型组小鼠的脾脏IL-6、IL-17、IL-18、IFN-γ在辐射后第1d、第14d与空白对照组相比均有不同程度的增加,而TLR4^(+/+)辐射模型组小鼠的IL-10和TGF-β1与空白对照组相比则有明显减少。TLR4^(+/+)中药组小鼠的IL-6和IL-10在辐射后第1d均较模型组明显恢复,IL-17、IL-18在第14d部分恢复,TGF-β1在第14d明显恢复。TLR4^(-/-)辐射模型组小鼠的各炎性细胞因子相对空白对照组亦有不同程度的变化,但TLR4^(-/-)中药组小鼠的炎性细胞因子表达相对辐射模型组无明显统计学意义。结论:补肾解毒方通过TLR4信号途径能显著减轻核辐射损伤,影响各炎性细胞因子的表达。Objective: To observe the effect of Bushen Jiedu decoction on the expression of inflammatory factors in the downstream of TLR4 (like receptor-4 Toll) signal transduction pathway in mice after radiation of 60^Coγ ray and to explore its mechanism. Methods: The C57BL/ 10J background mice ( TLR4^+/+) were randomly divided into TLR4^+/+ blank control group, TLR4+/+ radiation model group, TLR4^+/+ TCM group, under the same background TLR4 gene knockout mice (TLR4^-/-) were randomly divided into TLR4^-/- blank control group, TLR4^-/- radiation model group, TLR4^-/- TCM group. In addition to the two control groups, the rest of the four groups of mice were one-time accepted 6 Gy 60^Coγ-ray dose total body irradiation. The survival of mice after radiation in each group was observed? and the values of IL-6、 IL-10、 IL-17、 IL-18、 IFN_7、 TGF-pi in the spleen at 1 ,14and 30days were recorded. Results : IL-6 , IL- 17,IL-18, IFN-7 of the TLR4^+/+ radiation model group mice increased in different degrees(P〈0. 01) and IL-10 和 TGF- β1 significantly decreased compared with the blank control group at 1 ,14and 30days after irradiation. Compared with the model group,IL-6 and IL-10 of the TLR4^+/+ TCM group restored obviously at 1 day, IL-17、 IL-18 restored partially and TGF-β1 restored obviousl at 14 days. Inflammatory cytokines of the TLR4^-/- radiation model group were changed in different degree compared with the blank control group. Inflammatory cytokines had no statistical significance between the TLR4^-/- TCM group and the radiation model group. Conclusion: Bushen Jiedu decoction can significantly reduce the radiation injury and affect the expression of inflammatory cytokines through the TLR4 signal pathway. It has an ideal radiation protection function.
