摘 要:为了掌握浙江省近几年副猪嗜血杆菌(Haemophilus parasuis,HPs)的血清型流行情况,通过PCR扩增的方法来鉴定浙江省2010—2015年从113个猪场中分离鉴定的副猪嗜血杆菌血清型,同时检测6个可能与副猪嗜血杆菌毒力相关的基因(B14、D45、nha C、fhu A、hhd A、hhd B),并分析其与血清型的相关性。结果显示:浙江省副猪嗜血杆菌近几年的优势血清型为5/12型、4型、13型和14型,未检测到3型、9型、11型和15型;2010—2015年副猪嗜血杆菌的主要血清型流行趋势并未发生明显改变,部分猪场存在2种血清型菌株共感染现象;对潜在毒力基因的检测结果表明,D45基因在毒力菌株中出现的比例(82.4%~87.5%)高于其在无毒力菌株中的检测比例(50%);fhu A基因在强毒力菌株中的检测比例(55%)却低于其在中等毒力和无毒力菌株中的检测比例(70.6%~90%);其他4个毒力基因(B14、nha C、hhd A、hhd B)在所有菌株中均有较高的比例。提示:副猪嗜血杆菌潜在的毒力基因与血清型之间并没有明显的相关性。To investigate serovars of Haemophilus parasuis( HPs) in Zhejiang province,multiplex PCR was used to identify the serotype of HPs strains isolated from 113 farms during 2010- 2015 in Zhejiang. Meanwhile,six potential virulence- associated genes( B14,D45,nha C,fhu A,hhd A,hhd B) in these strains were detected,and then the correlation of the six genes with serotypes was analyzed. The results showed that 5 /12,4,13 and 14 were the major prevalent serotypes in Zhejiang and no isolates of serovars 3,9,11 and 15 were detected;the primary serotype prevalence during 2010- 2015 did not change significantly and the co- infection of two serotypes were detected in several farms. The detection results of potential virulence genes showed that the ratio of D45 gene in virulent strains( 82. 4%- 87. 5%) was higher than that in the avirulent strains( 50%); the proportion of virulent strains( 55%) harboring fhu A gene was lower than that of moderate virulent and avirulent strains( 70. 6%- 90%); the testing rates of the remaining four genes( B14,nha C,hhd A,hhd B) were high in all strains. The results indicated that there was no clear correlation between the six potential virulence- associated genes and serotypes.
