摘 要:目的分析哈萨克族肥胖儿童HFA小鼠的肠道菌群环境,以探索肠道菌群与哈萨克族儿童发生肥胖的关系。方法采用PCR-DGGE技术对20只哈萨克族肥胖儿童HFA小鼠模型进行检测,并对DGGE图谱上的优势条带回收、克隆并测序,对测序结果进行BLAST比对分析。结果各样本的DGGE图谱上均形成复杂而密集的电泳条带,含有大量的共同条带,主要有6种共同优势菌属序列:Bacteroidetes、Firmicutes、Lactobacillus、Escherichia、Bifidobacterium、Lactococcus。结论PCR-DGGE技术可以较好的呈现哈萨克族肥胖儿童HFA小鼠肠道内的主要菌属序列,为进一步探索哈萨克族儿童肥胖与肠道菌群关系的研究开辟了一条更为有效的途径。Objective To analysis intestinal flora environment of HFA mice related with Kazakh obesity children,in order to explore the relationship between intestinal flora and kazak children obesity.Methods PCR-DGGE method was adopted to detect the samples of HFA mice,the advantage stripe tags on DGGE profiles were recycled,cloned and sequenced,and BLAST results were compared.Results PCR-DGGE fingerprint showed each samples form complex and diverse species stripe,contained a lot of same bands.There were mainly six kinds of common advantage bacterium sequences:Bacteroidetes,Firmicutes,Lactobacillus,Escherichia,Bifidobacterium,Lactococcus.Conclusion The PCR-DGGE technology can present the intestinal sequence of main species in HFA mice of Kazakh obesity children,it has opened up a new effective way to explore the relationship between obesity and intestinal flora.
