摘 要:目的观察杜仲-牛膝配伍对卵巢切除所致实验性骨质疏松大鼠的血清雌二醇(E2)和股骨骨密度(BMD)的影响,探讨杜仲-牛膝药对防治绝经后骨质疏松(PMOP)的作用机制。方法取72只雌性SD大鼠随机分为假手术组、模型组、阳性对照组、杜仲组、牛膝组、杜仲-牛膝组共6组,每组12只。行卵巢切除术造模,其中假手术组只切除卵巢周围少量脂肪。造模成功后正常饲养7 d后开始给药,连续给药3个月后取腹主动脉血测定E2,并分离左侧股骨,检测BMD。结果与模型组比较,各给药组E2、BMD均显著增高(P<0.05),且杜仲-牛膝组E2水平显著高于杜仲组、牛膝组。结论杜仲-牛膝药对可显著提高去卵巢骨质疏松大鼠E2水平和股骨BMD,显示了其对PMOP的治疗效果,也为开发治疗PMOP的中药复方提供了依据。Objective To determine the effect of eucommia ulmoldes and achyranthes combination on the serum estrogen and femoral bone mineral density in ovariectomy induced osteoporosis rats, and to explore the therapeutic mechanism of eucommia ulmoldes and achyranthes on postmenopausal osteoporosis(PMOP). Methods Totally 72 female SD rats were divided into 6 groups 12 in each: a sham operation group, a model group, a positive control group, a eucommia ulmoldes group, an achyranthes group and a eucommia ulmoldes coupled with achyranthes group. A small amount of fat was cut in rats in the sham operation group around the ovary excision. Other groups were ovariectomized. After success modeling, the rats were fed normally intragastrically. They were administratered a total of 3 months. Blood was drawn from ventral aorta and the left side femur head isolated in each rat to determine serum estradiol and bone mineral density. Results Compared with the model group, the serum estradiol level and bone mineral density of each medication group were both increased significantly(P 0.05). The serum estradiol level of eucommia ulmoldes coupled with achyranthes group was significantly higher than that of eucommia ulmoldes group and achyranthes group. Conclusion The combination of eucommia ulmoldes and achyranthes can significantly increase the serum estradiol level and bone mineral density in PMOP rats, which indicates that they have good therapeutic effect on PMOP. This research provides an experimental reference for the development of Chinese medicine for PMOP.
