摘  要:目的:观察人工虫草多糖及核苷对D-半乳糖诱导大鼠衰老模型的治疗作用。方法:将40只SD雄性大鼠随机分为空白组、模型组、人工虫草多糖组与人工虫草核苷组,每组10只。除空白组大鼠颈背部皮下注射与模型组等容量的0.9%氯化钠注射液外,其余各组大鼠均采用颈背部皮下注射D-半乳糖法建立衰老模型。从注射的第12天开始,人工虫草多糖组与人工虫草核苷组分别灌服人工虫草多糖与人工虫草核苷溶液,空白组与模型组均灌服同体积0.9%氯化钠注射液,连续给药30d。结果:与模型组比较,人工虫草多糖及人工虫草核苷组的一般情况改善,体质量及胸腺、脾组织、脑指数提高,并提高了血液中SOD(超氧化物歧化酶)、GSH-PX(谷胱甘肽过氧化物酶)水平,且显著降低了MDA(丙二醛)水平(P<0.05,P<0.01),同时降低了心、脑及肝匀浆LPO(过氧化脂质)水平(P<0.01)。结论:人工虫草多糖及人工虫草核苷均能通过抗氧化作用,起到良好的抗衰老作用。Objective: To investigate the therapeutic effect of polysaccharides and nucleosides from cultured Cordyceps sinensis on D- galactose- induced aging rats. Methods: A total of 40 male Sprague- Dawley rats were divided into blank group,model group,Cordyceps sinensis polysaccharide group,and Cordyceps sinensis nucleoside group,with 10 rats in each group. The rats in the model group,Cordyceps sinensis polysaccharide group,and Cordyceps sinensis nucleoside group were treated with subcutaneous injection of D- galactose at the back of the neck to establish a model of aging,and those in the blank group were treated with subcutaneous injection of 0. 9% sodium chloride injection of the same volume as the model group at the back of the neck. Since day 12 of injection,the Cordyceps sinensis polysaccharide group or Cordyceps sinensis nucleoside group were treated with Cordyceps sinensis polysaccharide or nucleoside solution by gavage,and the blank group and the model group were treated with 0. 9% sodium chloride of the same volume by gavage. All the groups were treated for 30 consecutive days. Results: Compared with the model group,the Cordyceps sinensis polysaccharide group and the Cordyceps sinensis nucleoside group showed a significant improvement in general status,significant increases in body mass index,thymus index,spleen index,brain index,and the levels of superoxide dismutase and glutathione peroxidase in blood,and significant reductions in malondialdehyde content the levels of lipid peroxide in the heart,brain,and liver homogenate( P〈0. 05 or P〈 0. 01). Conclusion: Polysaccharides and nucleosides from cultured Cordyceps sinensis have a good anti- aging effect due to their antioxidant capacity.
