摘 要:目的:测定西黄丸中水溶性总糖含量。方法:以蒽酮-硫酸比色法,葡萄糖为对照,测定波长626nm。本实验对显色剂的浓度和总糖提取方法进行了改进,设计了四个浓度的显色剂,以一定的体积加入显色,分别在线性范围内做标曲,从整体上选择出最佳的显色剂浓度;总糖的提取方法改用称重补重的回流方法,在一定程度上提高了该提取方法的重复性。结果:葡萄糖在2.0-24.0μg/mL范围内呈良好的线性关系,标准曲线回归方程为Y=0.0555X+0.0019,R^2=0.9996,Y为吸光度,X为浓度。平均回收率97.5%(n=6),RSD=1.21%。结论:此法稳定可行,结果准确,专属性好,可用于测定西黄丸中总糖含量。Objective:To establish the method for quantification of total sugar in XihuangWan. Methods Colorimetric method was adopted with glucose as reference. The detection wavelength was 626nm. Methods:The chromogenic agent concentration and extraction method was improved. Four concentration levels of chromogenic agent were evaluated for optimizing the reaction condition by plotting the calibration curve,and the best choice was made according to the value of correlation coefficient. Results: There was a good linear relationship with the sugar concentration level in the range of 2.00-24.00μg/mL. The standard curve equation was Y =0.055 5X+0.001 9,R^2=0.9996. The average recovery was 97.5% withRSDof 1.21% (n = 6). Conclusion:The Method is Stable;Accurate and Specific,and it Can be Used for the Determination of Total Sugar in Xihuang Wan.
