血府逐瘀胶囊对子宫内膜细胞炎症模型影响的研究

摘　　要：目的:探讨血府逐瘀胶囊对子宫内膜细胞炎症模型影响的实验研究。方法:通过酶消化法获得原代子宫内膜细胞,脂多糖(LPS)诱导炎症模型,后实验随机分为5组,正常对照组,模型组(100μg·m L^(-1)LPS),血府逐瘀胶囊低、中、高剂量组(10、30、100μg·m L^(-1)血府逐瘀胶囊)。MTT法检测各组细胞活力,酶联免疫吸附法检测各组细胞中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL^(-1)β)、IL-6、IL-8蛋白含量,免疫印迹法检测各组细胞中基质金属蛋白酶-9(MMP-9),环氧化二酶(COX-2)蛋白表达量及p38丝裂原活化蛋白激酶(p38 MAPK)磷酸化水平,反转录聚合酶连锁反应检测p38 MAPK mRNA含量。结果:与正常组比较,模型组中细胞活力下降,TNF-α、IL^(-1)β、IL-6、IL-8、MMP-9及COX-2蛋白表达量都显著提高(P<0.01),p38 MAPK磷酸化水平提高(P<0.01),p38 MAPK mRNA表达量显著提高(P<0.01);与模型组比较,血府逐瘀胶囊低、中、高剂量组中细胞活力下降,TNF-α、IL^(-1)β、IL-6、IL-8水平都显著降低(P<0.01),p38 MAPK mRNA表达量显著降低(P<0.01);血府逐瘀胶囊中、高剂量组中MMP-9及COX-2蛋白表达量及p38 MAPK磷酸化水平下调(P<0.01)。结论:血府逐瘀胶囊能显著抑制LPS诱导的子宫内膜细胞炎症,与降低炎症因子表达及p38 MAPK信号通路有关。Objective： To explore the effect of Xuefu Zhuyu Capsule on inflammation in endometrial cell induced by lipopolysaccharide（ LPS）. Methods： Primary endometrial cells were obtained by enzymatic digestion. The cases were randomly divided into normal control group,model control group（ 100 μg·m L-1LPS）,Xuefu Zhuyu Capsule low,medium and high dose groups（ 10,30,100 μg·m L-1Xuefu Zhuyu Capsule）. The cell viability was examed by MTT method. The content of tumor nuclear factor（ TNF- α）,interleukin-1 β（ IL-1β）,IL- 6 and IL- 8 were examed by enzyme linked immunosorbent assay. The phosphorylation of p38 mitogen- activated protein kinase（ p38 MAPK） and the expression of matrix metalloprotein- 9（ MMP- 9） and cyclooxygenase- 2（ COX- 2） was assayed by western blot. The expression of p38 MAPK mRNA was assayed by RT- PCR. Results：Compared with normal control group,the cell viability was decreased. The concentrations of TNF- α,IL-1β,IL- 6,IL- 8,MMP- 9 and COX- 2 protein,the phosphorylation of p38 MAPK and the expression of p38 MAPK mRNA were increased in model group（ P 〈 0. 01）. Compared with model control group,the cell viability was increased. The concentrations of TNF- α,IL-1β,IL- 6,IL- 8 and the expression of p38 MAPK mRNA were decreased in model group than those in Xuefu Zhuyu Capsule low,medium and high dose groups（ P 〈 0. 05）. The expressions of MMP- 9 and COX- 2 protein and the phosphorylation of p38 MAPK were decreased in model group than those in Xuefu Zhuyu capsule medium and high- dose group（ P 〈 0. 05）. Conclusion： These results suggested Xuefu Zhuyu Capsule could inhibit inflammation in endometrial cell induced by LPS via blocking p38 MAPK signal pathway and down- regulating the expression of inflammatory factor.
