补肺益肾方联合舒肺贴对慢性阻塞性肺疾病大鼠T淋巴细胞亚群的影响

摘　　要：目的:评价补肺益肾方联合舒肺贴对慢性阻塞性肺疾病(COPD)稳定期大鼠T淋巴细胞亚群的影响。方法:大鼠72只随机分为空白组、模型组、舒肺贴组、补肺益肾组、补肺益肾联合舒肺贴组(简称联合组)、氨茶碱组,12只/组。采用熏烟联合细菌感染法制备COPD稳定期大鼠模型。自第9周,各组分别以相应方案处理,第20周结束后取材。观察大鼠肺功能及外周血和支气管肺泡灌洗液(BALF)中CD_3^+、CD_4^+、CD_8^+、CD_4^+/CD_8^+的表达。结果:模型组外周血、BALF中CD_3^+、CD_4^+、CD_4^+/CD_8^+水平较空白组明显降低(P<0.01),CD_8^+水平明显升高(P<0.01)。与模型组相比,各治疗组外周血、BALF中CD_4^+、CD_4^+/CD_8^+水平升高(P<0.05,P<0.01),CD_8^+水平降低(P<0.05,P<0.01);联合组CD_3^+水平升高(P<0.05,P<0.01)。联合组外周血、BALF中CD_4^+、CD_4^+/CD_8^+较穴位贴敷组升高(P<0.05,P<0.01),CD_8^+下降(P<0.05);BALF中CD_4^+、CD_4^+/CD_8^+较补肺益肾组升高(P<0.05)。结论:补肺益肾联合舒肺贴可调节CD_3^+、CD_4^+、CD_4^+/CD_8^+表达,从而提高机体免疫功能,发挥治疗作用。Objective： To observe the influence of Bufei Yishen Granule combined with acupoint sticking therapy on T lymphocytes subpopulations of chronic obstructive pulmonary disease（ COPD） rats. Methods： Seventy- two Sprague- Dawley rats were randomly assigned into control,model,Bufei Yishen,Shufei Plaster,Bufei Yishen ＋ Shufei Plaster and aminophylline groups,with12 animals in each group. COPD rats were exposed to cigarette smoke and Klebsiella pneumoniae（ KP） for model establishment.From week 9 through 20,the rats were treated with corresponding treatment protocols. CD3＋,CD4＋,CD8＋and CD4＋/CD8＋in blood and bronchoalveolar lavage fluid（ BALF） were measured. Results： Compared with control group,CD3＋,CD4＋and CD4＋/CD8＋in blood and BALF in model group decreased（ P 〈 0. 01） while CD8＋increased（ P 〈 0. 01）. Compared with model group,CD4＋and CD4＋/CD8＋in blood and BALF of treatment groups increased and CD3＋in Bufei Yishen ＋ acupoint sticking group increased while CD8＋decreased（ P 〈 0. 05,P 〈 0. 01）. CD4＋and CD4＋/CD8＋in Bufei Yishen ＋ Shufei Plaster group increased compared with that of Shufei Plaster group（ P 〈 0. 05,P 〈 0. 01） while CD8＋decreased（ P 〈 0. 05）. CD4＋and CD4＋/CD8＋in BALF in Bufei Yishen ＋ Shufei Plaster group was higher than those of Bufei Yishen group（ P 〈 0. 05）. Conclusion： Bufei Yishen Granule combined with Shufei Plaster can improve the body immunity function through regulating T lymphocytes subpopulations.
