以“肾脑相济”理论为指导的电针疗法对围绝经期抑郁症模型大鼠行为学及下丘脑-垂体-肾上腺影响

摘　　要：目的:研究以"肾脑相济"理论为指导的电针疗法对围绝经期抑郁症大鼠行为学及下丘脑-垂体-肾上腺轴的影响,探究以"肾脑相济"为治疗原则的电针疗法对围绝经期抑郁症的作用机制。方法:60只大鼠随机分为空白组、模型组、假手术组、西药组和电针组,采用雌性SD大鼠双侧卵巢切除和慢性不可预见刺激结合造模。西药组采用盐酸氯米帕明灌胃,电针组采用电针刺激"百会""肾俞""三阴交"。连续治疗28 d,阴道涂片、抓力实验及糖水偏嗜实验结果,运用酶联免疫吸附法(ELISA法),测定血清促肾上腺皮质激素释放激素(CRH)、促肾上腺皮质激素(ACTH)、皮质醇(CORT)的含量。结果:与模型组相比,电针组及西药组抓力增加(P<0.05),糖水消耗率升高(P<0.05),电针组及西药组血清CRH、ACTH、CORT含量低于模型组(P<0.05)。结论:以"肾脑相济"理论为指导的电针疗法可以通过改善大鼠行为学及调节下丘脑-垂体-肾上腺轴对围绝经期抑郁症起到防治作用。Objective： To study the effect of electroacupuncture therapy on the behavior and hypothalamus pituitary adrenal axis in the perimenopausal depression rats and explore the mechanism of electroacupuncture therapy in the treatment of perimenopausal depression. Methods： Sixty rats were randomly divided into blank group,model group,sham operation group,western medicine group and EA group. The western medicine group was treated with clomipramine hydrochloride orally and the electroacupuncture group received electroacupuncture at Baihui（ DU20）,Shenshu（ BL23）,Sanyinjiao（ SP6）,continuous treatment for 28 days. The vaginal smear,grasping force experiment and sucrose preference of the experimental results were studied by using enzyme linked immunosorbent assay（ ELISA）. We measured the content of serum corticotropin releasing hormone（ CRH）,adrenal cortical hormone adrenocorticotropic hormone（ ACTH） and cortisol（ CORT） content. Results： Compared with the model group,electroacupuncture group and western medicine group’s grasping force increased（ P 〈 0. 05） and the sugar consumption rate increased（ P〈 0. 05）. The electroacupuncture group and western medicine group’s serum levels of CRH,ACTH and CORT were higher than those of the model group（ P 〈 0. 05）. Conclusion： The theory of harmony between kidney and brain theory is the guidance of electroacupuncture therapy can improve the behavior of rats and regulate the hypothalamus pituitary adrenal axis to prevent and treat the depression in peri- menopausal period.
