针康法对局灶性脑梗死大鼠神经功能缺损及梗死灶周边区RhoA、ROCK-Ⅱ蛋白表达的影响  被引量:9

摘  要:目的:探讨针康法对局灶性脑梗死大鼠神经功能缺损及梗死灶周边区Rho A、ROCK-II蛋白表达的影响。方法:将90只清洁级SD雄性大鼠随机分为5组,即假手术组、模型组、针刺组、康复组及针康组,各组于3天、7天、14天时间点再分成3个亚组(n=6)。采用改良的Zea Longa线栓法制备局灶性脑梗死大鼠模型。假手术组与模型组大鼠不予任何治疗,针刺组给予头穴丛刺治疗,康复组给予运动康复训练,针康组给予头穴丛刺结合运动康复训练。术后3天、7天、14天采用改良大鼠神经功能缺损评分(m NSS)评估各组大鼠神经功能,免疫组化法检测大鼠梗死灶周边区Rho A、ROCK-Ⅱ蛋白的表达。结果:术后7天、14天,与模型组比较,各干预组大鼠神经功能缺损评分均明显降低(P<0.05);与针刺组、康复组比较,针康组大鼠评分有所降低(P<0.05)。术后各时间点,与模型组比较,各干预组梗死灶周边区Rho A、ROCK-Ⅱ阳性细胞数均明显减少(P<0.05);与针刺组、康复组比较,针康组大鼠梗死灶周边区Rho A、ROCK-Ⅱ阳性细胞数目有所减少(P<0.05)。结论:头穴丛刺疗法、运动康复训练、针康法均能有效改善局灶性脑梗死大鼠神经功能缺损,下调梗死灶周边区Rho A、ROCK-Ⅱ蛋白表达,且针康法优于单纯头穴丛刺或运动康复训练。Objective: To investigate the effect of cluster needling of scalp acupuncture combined with rehabilitation on neurological function and expression of RhoA and ROCK- Ⅱ in the cortical peri- infarct area of rats after focal permanent cerebral ischemia. Methods: 90 male SD rats were randomly divided into five groups,namely,sham control group( SC),model group( M),scalp acupuncture group( SA),rehabilitation training group( RT) and acupuncture- rehabilitation therapy group( ART). Each group was subdivided into 3 subgroups by 3 d,7 d,14 d( n = 6). The rats in M,SA,RT,ART groups were made into the models with focal permanent cerebral ischemia by modified Zea Longa method. Rats in SC group and M group were without any intervention. SA group was treated by cluster needling of scalp acupuncture therapy; RT group was treated by rehabilitation training; ART group received acupuncture- rehabilitation therapy. Neurological function was evaluated by modified neurological severity score( m NSS),and the expressions of RhoA and ROCK-Ⅱ in the cortical peri- infarct area of rats were observed by immunohistochemistry staining in rats in each group after 3 d,7 d,14 d of surgery. Results: After 7 d,14 d of surgery,compared with SA group and RT group,the m NSS was reduced in ART group,and the difference was statistically significant( P〈0. 05). After 3 d,7 d,14 d of surgery,compared with SA group and RT group,the number of positive cells of RhoA and ROCK-Ⅱ was decreased in ART group,and the difference was statistically significant( P〈0. 05). Conclusion: Acupuncture- rehabilitation therapy can improve the neurological function defect in rats with focal permanent cerebral ischemia,and its therapeutic mechanism may be related to down- regulating the expressions of RhoA and ROCK-Ⅱ in the cortical peri- infarct area.
