摘　　要：目的:探讨头穴透刺对大脑中动脉局灶性脑缺血再灌注(MCAO/R)大鼠缺血侧脑微血管内皮细胞紧密连接蛋白ZO-1表达的影响。方法:130只SD雄性大鼠随机分为假手术组、模型组和头针组。采用改良的线栓法复制右侧大脑中动脉缺血2 h再灌注模型,针刺大鼠患侧百会透曲鬓穴;应用免疫组化和Western blot技术定性和半定量检测再灌注后不同时间点各组大鼠缺血侧脑组织中ZO-1蛋白的表达。结果:头针组大鼠在再灌注后1天、2天、3天,缺血侧脑组织中ZO-1蛋白含量均较模型组显著增加(P<0.05),但仍低于假手术组。结论:早期进行头穴透刺治疗能上调脑缺血再灌注后ZO-1蛋白表达,这可能是头穴透刺疗法减轻脑缺血再灌注后血脑屏障(BBB)结构破坏,抑制紧密连接(TJ)开放,保护BBB的机制之一。Objective： To investigate the effect of Baihui- Qubin scalp- acupuncture on the expression of ZO- 1 after cerebral ischemia / reperfusion. Methods： 130 adult male SD rats were randomly divided into a sham group,an ischemia / reperfusion（ I / R） group and a scalp- acupuncture group. Models were induced by focal ischemia for 2h with middle cerebral artery occlusion and reperfusion（ MCAO / R）. Rats in scalp- acupuncture group were acupunctured at the trouble side by Baihui- Qubin scalp- acupuncture. We detected the positive expression and protein contents of ZO- 1 by immunohistochemistry and the western blot method respectively in brain tissue of each set. Results： Compared with model group,the brain ZO- 1 protein contents had been significantly increased in the scalp- acupuncture group（ P〈0. 05）. Conclusion： Baihui- Qubin scalp- acupuncture can protect the blood brain- barrier from I / R injury. Its mechanism may be related to up- regulating the expression of ZO- 1 protein.
