摘 要:目的:考察茜草炭各极性部位对子宫出血模型大鼠的止血作用。方法:空白组与其余各组分别灌胃给予0.5%羧甲基纤维素钠(CMC-Na)、戊酸雌二醇(2.5 mg·kg^(-1)),连续灌胃3周,以宫血宁为阳性药,对照组及模型组灌胃(ig)CMC-Na,阳性组、石油醚提取物组、乙酸乙酯提取物组、正丁醇提取物组和萃取剩余部位组分别ig相应的治疗药物,观察茜草炭各极性部位对子宫出血模型大鼠血液流变学、组织形态学、血小板系统和凝血系统的影响。结果:与空白组比较,模型组大鼠各切变率下的全血黏度降低(P<0.01),TT、PT、APTT延长(P<0.05、0.01),FIB、6-keto-PGF1α含量与ADP%升高(P<0.01),6-keto-PGF1α/TXB2比值升高(P<0.01),TXB2含量降低(P<0.01)。与模型组比较,乙酸乙酯提取物组和正丁醇提取物组能显著改善模型大鼠全血黏度(P<0.05,P<0.01),缩短TT、PT、APTT(P<0.05,P<0.01),减低FIB、6-keto-PGF1α含量(P<0.05,P<0.01),乙酸乙酯提取物组显著升高TXB2含量(P<0.05);石油醚提取物组和萃取剩余部位组均与ADP%具有改善模型大鼠全血黏度、缩短TT、APTT与PT,降低FIB和ADP%,降低TXB2含量与6-keto-PGF1α含量的趋势。结论:茜草炭乙酸乙酯提取物和石油醚提取物主要通过降低全血黏度、改善凝血功能以及促进血小板聚集功能发挥止血作用,为茜草炭止血有效部位。Objective: To discuss the hemostasis of the different polarities of Rubiae Radix et Rhizoma (carbonized) on the dysfunctional uterine bleeding rat model. Methods: The SD rats were divided into seven groups. The rats in the control group were fed with CMC-Na for 3 weeks, the other groups were fed with estradiol valerate for 3 weeks. Gong Xue Ning was taken as the positive control drug. The rats in the control group and model group were fed with CMC-Na for 7 days, and the rats in other groups were fed with corresponding drugs simultaneously. The influence of the different polarities of carbonized Rubia cordifolia compound to dyfunctional uterine bleeding (DUB) rat's hemorheology, coagulation system, fibrinolytic system, tissue morphology and platelet system. Results: Compared with the control group, the model group could reduce the whole b-lood viscosity in each shear rate (P 〈 0. 01 ) and prolong the TT, PT, APTT( P 〈 0. 05, P 〈 0.01 ), increase the FIB, 6- keto-PGFlα content and ADP% (P 〈 0. 01 ), and decrease the TXB2. Compared with the model group, ethyl acetate and butanol extract group could increase the whole b--lood viscosity in each shear rates ( P 〈 0. 05, P 〈 0. 01 ), shorten the TT, PT, APTT(P〈0.05, P〈0.01), decrease the content of FIB, 6-keto-PGF1α and ADP% (P 〈0.05, P 〈0.01), and increase the TXB2 , while petroleum ether and water extract groups gave no significant effect. Conclusion: Ethyl acetate extract is the active part of Rubiae Radix et Rhizoma (carbonized), showing the effect of hemostasis by reducing the low whole blood viscosity, improving coagulation function mainly and ameliorating the function of platelet aggregation.
