摘 要:为探索南天竹种质资源离体保存的方法,以南天竹试管苗为材料,研究不同浓度ABA和CCC对其离体保存时的生长、增殖、存活率及恢复生长的影响。结果表明,CCC对株高的抑制大于ABA,对增殖的抑制较ABA弱,最高存活率(83.33%)远大于ABA的(56.67%)。适合南天竹试管苗离体保存的培养基为MS+30 g·L^(-1)蔗糖+0.5 mg·L-16-BA+0.05 mg·L^(-1)NAA+5.0 mg·L^(-1) CCC,保存120d后,存活率为83.33%、增殖系数为2.33、株高为1.59 cm,恢复生长成活率达96.67%,且形态正常。添加CCC可延缓南天竹试管苗生长,用于南天竹种质资源的离体保存。The effects of different concentration of ABA and CCC on the growth, proliferation, survival rate and recovery growth of test-tube shoots during in vitro preservation were studied for exploring a method for in vitro preservation of Nandina domestica. The resuhs showed that the inhibition degree of plant height by CCC treatment was deeper than ABA treatment, the inhibition effect on the proliferation was reversed, and the survival rate(83.33% )by CCC treatment was obviously higher than ABA treatment (56. 67% ). So the suitable medium for N. domestica in vitro preservation was MS + 30 g.L^-1 sucrose + 0. 50 mg-L^-1 6 - BA +0. 05 mg·L^-1NAA + 5 mg·L^-1 CCC, on which in vitro preservation for 120 d, the survival rate, proliferation coefficient and plant height was 83.33%, 2. 33 and 1.59 cm, respectively. After in vitro preservation for 120 d, the survival rate on recovery culture reached 96. 67%, and their morphology was normal. By adding CCC, the growth of test-tube shoots was alleviated which could applicate for N. domestica germplasm in vitro preservation.
