摘 要:目的:观察川芎嗪防治急性肾缺血再灌注损伤(RIRI)的作用,探讨其损伤保护作用的可能机制。方法:选择SD大鼠50只,随机分成假手术组、对照组、川芎嗪低、中、高剂量组,每组各10只。各组分别于术前和术后24 h取血标本,检测血清肌酐(SCr)、尿素氮(BUN)含量;各组于术后24 h再次麻醉,摘取左肾,作组织学分析及检测组织匀浆中SOD、MDA和TNF-α含量。结果:与对照组比较,川芎嗪低、中、高剂量组肾组织学评分、血清SCr、BUN含量以及肾组织MDA和TNF-α水平明显降低,SOD活性升高,并呈剂量依赖关系。结论:川芎嗪对急性肾缺血再灌注损伤具有保护作用,提高肾组织SOD活性、降低MDA和TNF-α水平是其减轻损伤的可能机制。Objective:To observe the effect of ligustrazine on acute renal ischemia reperfusion injury of rats and investigate its mechanisms of protective effect on renal injury. Methods:50 SD rats were randomly divided into sham operation group, control group and ligustrazine group of low,medium and high dosage. Blood samples were taken before operation and 24 h after operation respectively,and contents of serum creatinine (SCr),urea nitrogen (BUN)were detected. All the rats were anaesthetized 24 h after operation again,and left kidney was removed,then the contents of SOD,MDA and TNF-α in tissue homogenate were detected and histological analysis was performed. Results:Compared with control group,contents of SCr,BUN,MDA and TNF-α and renal histology score were decreased and SOD activity was increased in ligustrazine groups of low,medium and high dosage with positive dose-dependent relationship. Conclusion:Ligustrazine has protective effect on acute renal ischemia reperfusion injury of rats through improving SOD activity of renal tissue and decreasing its contents of MDA and TNF-α.
