摘 要:目的:研究温胆汤对精神分裂症模型鼠海马组织神经调节蛋白1(NRG1)、Erb B4蛋白表达的影响。方法:将50只大鼠随机分为5组,即正常组、模型组、温胆汤40g/kg组、温胆汤20g/kg组、温胆汤10g/kg组。正常组和模型组分别用20 ml/kg的生理盐水ig,温胆汤按40,20,10 g/kg ig 20 ml/kg,1次/d,共21 d。在最后1次ig给药2 h后,分别对模型组及温胆汤组1次性ip MK-801 0.6 mg/kg以诱发精神分裂症模型,正常组ip给予等容积的生理盐水。造模后观察并记录各组大鼠的行为学改变,5 d后,处死并取其海马组织待测。选用MORRIS水迷宫测试大鼠学习记忆功能,免疫组化检测海马组织NRG1活性的表达,Western blot测定海马组织NRG1、Erb B4蛋白表达。结果:与正常组比较,模型组大鼠在造模后第2、第3天定位航行实验平均逃避潜伏期明显延长,空间探索实验穿越原平台位置次数和原平台位置所在象限停留时间减少;海马组织NRG1活性表达和NRG1、Erb B4蛋白表达水平显著升高。与模型组比较,温胆汤40、20、10 g/kg组均能降低定位航行实验平均逃避潜伏期,温胆汤40、20 g/kg组明显降低海马组织NRG1活性表达,温胆汤40、20、10 g/kg组能降低NRG1、Erb B4蛋白表达水平;温胆汤40、20 g/kg组增加空间探索实验穿越原平台位置次数和原平台位置所在象限停留时间。结论:温胆汤通过降低精神分裂症易感基因NRG1及其受体Erb B4蛋白的表达,改善大鼠的学习记忆功能,从而防治精神分裂症。Objective:To research the effect of Wendan Tang on neuregulin-1 ( NRG1 ) and ErbB4 of hippocampus in rats with schizophrenia. Method : Fifty SD rats were randomly divided five groups : normal group, model group, Wendan Tang 40 g/kg group, Wendan Tang 20 g/kg group and Wendan Tang 10 g/kg group. The rats of normal group and model group were given normal saline, The rats of Wendan Tang 40, 20,10 g/kg groups were respectively given Wendan Tang 40,20,10 g/kg,once a day,which last for 21 days. At the last given Wendan Tang or saline after two hours, the model group and Wendan Tang 40,20,10 g/kg groups were ip given MK-801 0.6 mg/kg to establish rat schizo- phrenia model, and the change of stereotyped behavior of rats were observed and recorded. After five days, the rats were sacrificed,the hippocampus tissue were tested. The learning and memory function of rats were tested by Morris water maze. The activity expression of NRG1 was detected by immunohistochemical method. The expression of NRG1 and ErbB4 were determined by Western bloting method. Result: Compared with the normal group, average escape latency of place navigation test was significantly longer ( P 〈 0.01 ), across the former platform position number and original quadrant platform position of spatial probe were less (P 〈 0.05 ~ P 〈 0.01 ), the activity expression of NRG1 and the expression of NRG1 and ErbB4 in hippocampal tissue were significantly increased (P 〈0.05 ~ P 〈0.01 ) in rats of the model group on the second and third days after modeling. Compared with the model group, Wendan Tang 40, 20, 10 g/kg groups could reduce average escape latency of place navigation test (P 〈 0.05 ~ P 〈 0.01 ). Wendan Tang 40, 20 g/kg groups could significantly reduce the activity expression of NRGI in hippocampal tissue (P 〈0.01 ). Wendan Tang 40, 20, 10 g/kg groups could reduce the expression of NRG1, ErbB4 in hipp- ocampal tissue ( P 〈 0.05 ~ P 〈 0.01 ). Wendan Tang 40, 20 g/kg groups could increase across the form
